Embodiment 1

[0034] Weigh 50 mg of dexamethasone with an electronic balance, and dissolve it in 10 ml of olive oil, stir and shake to dissolve completely to obtain an oil phase solution of 5 mg per ml. Dissolve 0.25ml of TWEEN-20 in 50ml of ultrapure water and stir well. Weigh 6 grams of polyvinyl alcohol (PVA) with an electronic balance, dissolve it in ultrapure water, stir and vibrate to dissolve completely, and obtain an aqueous phase solution. Under the condition of stirring, 2.5 mL of the oil phase solution was added dropwise into the polyvinyl alcohol aqueous solution, and the stirring was continued until a uniform emulsion was obtained. The obtained emulsion was electrospun by using a needle-free electrospinning device, the distance between the spinneret and the collecting drum was 15 cm, the applied voltage was 60 kV, and the spinneret speed was 5 revolutions per minute. After one hour, 4.12 g of a three-dimensional nanofiber mat with a thickness of about 0.4 mm and a nanofiber di...

Embodiment 2

[0036]Weighed 30 mg of paclitaxel with an electronic balance, and dissolved it in 10 ml of olive oil, stirred and oscillated to dissolve completely to obtain an oil phase solution of 3 mg per ml. Dissolve 0.25ml of TWEEN-20 in 50ml of ultrapure water and stir well. Dissolve 6.5 g of polyvinyl alcohol (PVA) in ultrapure water with an electronic balance, stir and vibrate to dissolve completely, and obtain an aqueous phase solution. Under the condition of stirring, 2.5 mL of the oil phase solution was added dropwise into the polyvinyl alcohol aqueous solution, and the stirring was continued until a uniform emulsion was obtained. The obtained emulsion was electrospun by using a needle-free electrospinning device, the distance between the spinneret and the collecting drum was 18 cm, the applied voltage was 65 kV, and the spinneret speed was 5 revolutions per minute. After one hour, 3.54 grams of a three-dimensional nanofiber mat with a thickness of about 0.4 mm was obtained on the...

Abstract

The invention relates to a method for preparing skin-core structured drug-loading nanofiber through a needleless electrostatic spinning technology, which comprises the following steps of: dissolving fat soluble drugs into vegetable oil to obtain oil-phase solution; dissolving water-soluble polymer materials into water, and then adding an emulsifier to obtain aqueous solution, wherein the use amount of the emulsifier is 0.1-5% of the total weight of the solution; adding the oil-phase solution into the aqueous solution drop by drop under a constant-speed stirring condition to obtain oil-in-water type O/W emulsion; and electrostatically spinning the emulsion on needleless electrostatic spinning equipment to obtain the skin-core structured drug-loading nanofiber. According to the preparation method disclosed by the invention, the operation is simple, the cost is low, economic benefits can be obtained, the production rate can be up to 8g of nanofiber per hour; furthermore, the prepared skin-core structured nanofiber has high mechanical property; and loading of fat soluble drugs is realized.

Description

technical field [0001] The invention belongs to the field of preparation of drug-loaded nanofibers, in particular to a method for preparing skin-core drug-loaded nanofibers by using needle-free electrostatic spinning technology. Background technique [0002] Electrospinning means that under the action of a high-voltage electric field, the polymer solution will be stretched by the electric field force to form a Taylor cone. As the voltage increases, the electric field force will overcome the surface tension of the solution, and the polymer solution will be drawn and thinned from the top of the Taylor cone, and sprayed to the receiving device of the ground wire at an accelerated rate. During electrospinning, the solvent evaporates and the nanofibers can be collected on a receiving device. In simple terms, the electrospinning device includes an electrostatic generator, a spinneret and a receiving device, and the solution is ejected from the spinneret by applying a high voltage...
