Embodiment 1

[0027] Example 1 Isolation and identification of Brevibacterium lateralis N106-2

[0028] Sampling time: August 2013

[0029] Sampling location: Taiyuan City, Shanxi Province

[0030] Using the gradient dilution method, 10 g of the obtained weed rhizosphere soil was added to a 90 mL sterile water conical flask, shaken at 150 rpm for 30 min to prepare a suspension, and then 1 mL of the suspension was taken and placed in a 9 mL sterile water conical flask, Prepare its 10 in sterile water 5 , 10 6Take 330ul of the diluted solution in each of them, apply LB medium plate, repeat each treatment 3 times, place the above medium at 28°C for constant temperature cultivation for 2d, pick a single colony and transfer it to LB medium for streak purification, purify The strains were stored in glycerol at -70°C.

[0031] The applicant named the isolated strain as N106-2, and its cultured form on LB medium is as follows: figure 1 As shown, the edge of the colony is smooth, gray-white, th...

Embodiment 2

[0034] Example 2 Broad-spectrum antibacterial test of Brevibacterium terpenoids N106-2

[0035] The Brevibacillus terpenoids N106-2 obtained in Example 1 was activated on LB medium, then transferred to 50 mL of LB medium, and cultured with shaking at 150 rpm at 28°C for 2 d, for use, with a bacterial content of 1 × 10 8 cfu / mL.

[0036] (1) PDA of plant pathogenic fungi (respectively: tomato fusarium wilt, capsicum root rot Fusarium solani, tomato botrytis cinerea, rice sheath blight, cabbage black spot, pear anthracnose and pear rot) PDA The cultured bacterial cake (7mm in diameter) is placed on one side of the PDA plate (9cm in diameter), the bacterial cake is 30mm away from the edge of the plate, and on the other side of the plate, 30mm away from the edge of the plate, and 5mm sterilization filter paper is placed in parallel, sterilization filter paper 15mm from the center of the plate, add 10 μL of the above-prepared Bacillus terpenoids N106-2 bacterial solution to the fi...

Embodiment 3

[0042] Example 3 Detection of antibacterial related substances of Brevibacillus lateralis N106-2

[0043] Brevibacillus lateralis N106-2 obtained in Example 1 was activated on LB medium, then transferred to 50 mL of LB medium, and cultured with shaking at 150 rpm at 28°C for 2 d, for use, with a bacterial content of 1 × 10 8 cfu / mL.

[0044] (a) Protease detection: Place a 5mm sterilized filter paper in the center of the protease detection medium plate, drop 5 μL of Brevibacillus terpenoids N106-2 bacterial solution on each filter paper, repeat 4 dishes for each treatment, and culture at 30°C for 2 days. Transparent circles are generated.

[0045] (b) Chitinase detection: Place a 5mm sterilized filter paper in the center of the chitinase detection medium, drop 5 μL of Brevibacillus lateralis N106-2 bacterial solution on each filter paper, repeat 4 dishes for each treatment, and culture at 30°C 3 ~ 7d, observe whether there is a transparent circle.

Abstract

The invention discloses a Brevibacillus laterosporus strain N106-2 of which the collection number is CGMCC No.8482. The strain has strong indoor flat inhibiting actions on multiple plant pathogenic fungi (fussarium wilt of tomato, root rot of chili, gray mold of tomato, Rhizoctonia solani, black spot of cabbage, anthracnose of pear and black rot of pear) and plant pathogenic bacteria (bacterial wilt of tomato, fire blight of pear and streak of rice); and the strain can secrete proteinase, cellulase and surfactant antimicrobial related substances with hemolytic activity. The Brevibacillus laterosporus can be prepared into a biocontrol aqua; the biocontrol aqua has certain preventing and treating effects on fussarium wilt of tomato, fusarium rot of chili, anthracnose of pome and eyespot of pome. The Brevibacillus laterosporus N106-2 has the advantages of no toxicity, no pathogenicity and high safety for human and livestock, is environment-friendly, is a useful resource for development of microbial pesticides, and has potential application prospects.

Description

technical field [0001] The invention relates to the field of biological pesticides, in particular to the research and development of a strain of Brevibacillus laterosporus and its application. Background technique [0002] For nearly half a century, chemical pesticides have played an important role in the prevention and control of agricultural pests and diseases, but the unreasonable use of pesticides and long-term and large-dose use have led to the problem of pesticide residues, which seriously endangers environmental safety. The use of antagonistic microorganisms to control plant diseases began in 1921. After years of development, microbial preparations that can produce active substances or biological pesticides containing active ingredients have become the hotspot and focus of research in recent years. At present, in the use of pesticides in my country, Biopesticides only account for 1%, and there are few varieties. Therefore, based on the background of sustainable agricul...
