Embodiment 1

[0022] The preparation of embodiment 1 high-efficiency phosphorus-dissolving and growth-promoting microbial bacterial agents

[0023] A fungus A44 was screened from the garlic farmland soil (calcareous soil) in Jining City, Shandong Province, my country, which has a strong effect of dissolving insoluble phosphorus in soil. Based on the observation of the morphological characteristics of the strain and the results of molecular biology sequencing, the strain A44 was finally determined to be Aspergillus niger.

[0024] 1. Cultivation of Aspergillus niger A44

[0025] (1) Composition of medium (kg / t fermentation liquid)

[0026] Soluble starch 10kg, soybean cake powder 10kg, corn flour 5kg, glucose 1kg, sucrose 10kg, yeast powder 5kg, (NH 4 ) 2 SO 4 3kg,K 2 HPO 4 0.2kg, NaCl2.5kg, MgSO 4 ·7H 2 O0.1kg, CaCO 3 0.5kg, FeSO 4 0.001kg, boric acid (1%) 1L, pH7.0, prepared with water.

[0027] (2) Fermentation culture

[0028]Inoculate Aspergillus niger A44 on potato sucrose (...

Embodiment 2

[0033] Embodiment 2 solid plate test phosphorus dissolution effect

[0034] In order to clarify the ability of Aspergillus niger A44 to dissolve various insoluble phosphorus, 5 phosphorus sources were used in this example, namely tricalcium phosphate Ca 3 (PO 4 ) 2 , iron phosphate, aluminum phosphate, zinc phosphate, phosphate rock powder and other phosphorus sources, cultured at 28°C for 5 days, and determined the available phosphorus on the PDA plate. In the experiment, Penicillium bailei ATCC20851 was used as the control strain, and a control experiment was set up.

[0035] The results showed that the A44 fungus had the strongest ability to dissolve tricalcium phosphate, and its phosphorus solubility was as high as 58.16 mg phosphorus / plate, which was higher than that of the phosphorus-dissolving Penicillium oxalicum P8 disclosed in ZL200810118076.9, and its phosphorus solubility was 48.76 mg phosphorus / plate , and higher than the phosphorus-dissolving capacity of strai...

Embodiment 3

[0039] The effect of embodiment 3 phosphate-dissolving bacterial strain A44 dissolving insoluble phosphorus in liquid medium

[0040] Under the condition of liquid culture, the experiment of dissolving ability of phosphate-dissolving fungus A44 to phosphate rock powder, aluminum phosphate, iron phosphate, calcium phosphate and zinc phosphate was carried out. The results showed that the available phosphorus dissolved by Aspergillus niger A44 from Yunnan phosphate rock powder reached 411.1 μg phosphorus / mL, accounting for 56.4% of the phosphorus content in the added phosphate rock powder; the available phosphorus released by Aspergillus niger A44 strain from tricalcium phosphate was the highest, Up to 815.2 μg phosphorus / mL, accounting for 82.1% of the added amount; bacterial strain A44 has the best ability to dissolve aluminum phosphate, and the available phosphorus is 411.1 μg phosphorus / mL, accounting for 32.4% of the added phosphorus amount (1270 μg phosphorus / mL); A44 has a...

Abstract

The invention discloses an efficient phosphorus-dissolving growth promoting bacterium-aspergillus niger A44 which is screened from calcareous soil in northern China, and provides a microorganism bacterium agent and bio-organic fertilizer prepared from the strain A44. The strain can efficiently dissolve phosphate rock powder, iron phosphate, aluminium phosphate, zinc phosphate and tricalcium phosphate, the phosphorus-dissolving kinds are more, and the phosphorus-dissolving capacity is high and exceeds that of internationally-recognized phosphorus-dissolving penicillium bilaii reported at present. The A44 microorganism bacterium agent can be compounded with sterile rotten livestock manure fertilizer, straw rotten matter, a binding agent and the like to form the bio-organic fertilizer. According to the bacterium agent and the bio-organic fertilizer thereof, the crop growth is significantly promoted, and compared with a control group, the corn biomass is increased by 80.9%, the peanut biomass is increased by 20.51%-24.14%, and the sunflower biomass is increased by 15.5%-32.3%; when the bacterium agent and the bio-organic fertilizer thereof are used in middle saline-alkali farmlands, the yield of sunflowers is increased by 33.3%; the growth promoting and yield increasing effects are not influenced by the used fertilizer, and the bacterium agent and the bio-organic fertilizer thereof can be applied on the condition that the fertilizer is normally used; the important significance on phosphate fertilizer resource saving, sustainable grain yield increasing and farmland ecology environment protecting of agricultural production in China is achieved.

Description

technical field [0001] The invention belongs to the field of microbiology, and in particular relates to a high-efficiency phosphorus-dissolving and growth-promoting bacterium, a bio-organic fertilizer prepared therefrom and its application. Background technique [0002] Phosphorus is the second most important element for plant growth after nitrogen, but its availability is very low due to soil fixation, making it difficult to meet the needs of crop growth (Wild, 1988). Studies have shown that more than 90% of the phosphorus fertilizer applied to the soil is converted into insoluble phosphorus, acidic soil is converted into iron phosphate and aluminum phosphate, and calcareous soil is converted into calcium phosphate (Hagin and Hadas, 1962; NagelschmidtG, et al. 1994). In order to increase crop yield, the amount of phosphate fertilizer has increased sharply, and phosphate rock resources are facing a crisis of depletion. It is estimated that the world's phosphate rock can be ...
