Embodiment 1

[0034] Purchase and prepare media and reagents first:

[0035] Preparation method of Staphylococcus aureus selective plate:

[0036] Baird-Parker medium, potassium tellurite egg yolk enrichment solution and freeze-dried rabbit plasma were purchased from Beijing Luqiao.

[0037] Weigh 6.3g of Baird-Parker culture based on 95mL of distilled water and heat to boil until completely dissolved. After autoclaving at 121°C for 15 minutes, add 5mL of potassium tellurite egg yolk enrichment solution, cool to 50±1°C and add 0.5mL of freeze-dried rabbit Plasma, poured into the plate immediately after mixing.

[0038] The rest of the medium and reagents refer to GB 4789.10-2016 "National Food Safety Standard Food Microbiological Examination of Staphylococcus aureus".

[0039] The rapid detection method of Staphylococcus aureus in food and drink, comprises the following steps:

[0040] 1) Bacteria enrichment: Weigh 25g of the catering sample to be tested into 225mL of 7.5% sodium chlorid...

Embodiment 2

[0045] Purchase and prepare media and reagents first:

[0046] Same as Example 1

[0047] The rapid detection method of Staphylococcus aureus in catering, comprises the following steps:

[0048] 1) Bacteria enrichment: Weigh 25g of the catering sample to be tested into 225mL of 7.5% sodium chloride broth, homogenize at 8000r / min for 2min, and incubate anaerobically at 37°C for 20h;

[0049] 2) Isolation: inoculate the enriched culture on the selective plate of Staphylococcus aureus by streaking, and incubate anaerobically at 37°C for 30 hours;

[0050]3) Preliminary identification: Staphylococcus aureus is round on the Staphylococcus aureus selective plate, the surface is smooth and raised, the colony diameter is 2-3mm, the color is gray-black to black, shiny, often light-colored ( non-white) surrounded by an opaque ring (precipitation) with a clear transparent band outside.

[0051] 4) determine identification: step 3) pick the suspicious bacterium colony on the selective ...

Abstract

The invention provides a method for rapidly detecting staphylococcus aureus in diet. The method comprises the following steps: 1) culture enrichment: weighing a sample to sodium chloride broth, and carrying out anaerobic culture after homogenization; 2) separation: carrying out streak inoculation on a culture after culture enrichment to a staphylococcus aureus selective flat plate, and carrying out anaerobic culture; 3) preliminary identification: preliminarily determining a staphylococcus aureus bacterial colony according to morphology; and 4) confirmation identification: inoculating suspicious bacteria on the staphylococcus aureus selective flat plate on a blood plate, culturing, observing whether the suspicious bacteria are dissolved in blood, selecting bacterial colonies which are dissolved in blood and carrying out biochemical identification by using a VITEK 2 COMPACT full-automatic microorganism analyzing system to obtain the result. Compared with the prior art, the method has the advantages that by the characteristics that the staphylococcus aureus is aerobic and anaerobic bacteria, and the metabolites of the staphylococcus aureus can produce plasma-coagulase, the suspiciousbacteria which are not dissolved in blood do not need to be identified, the staphylococcus aureus detecting time is shortened, and the detecting difficulty is reduced.

Description

technical field [0001] The invention belongs to the field of food detection, and in particular relates to a rapid detection method for Staphylococcus aureus in catering. Background technique [0002] Catering is the basis for people's survival. With the acceleration of the pace of life, the increase of work pressure, especially the rise of Internet sales, people's demand for popular catering is showing an upward trend every year, and online ordering has become a popular trend. [0003] The routine detection items of pathogenic bacteria in catering food are Staphylococcus aureus and Salmonella. Staphylococcus aureus is a facultative anaerobic bacterium belonging to Micrococcus family. It has strong resistance and wide distribution. It is widely found in air, dust, water and human and animal excrement. It is one of the common pathogenic bacteria, and it is also the pathogenic bacteria with the highest detection rate in catering and food testing. [0004] According to the rep...
