Embodiment 1

[0094] 1. Acrylic modification of polylactic acid-caprolactone (P(CL-DLLA))

[0095] The acrylic acid modified scaffold material was prepared by ultraviolet grafting method, and the specific operation was as follows:

[0096] (1) Soak the polylactic acid-caprolactone (P(CL-DLLA)) scaffold material in a hydrogen peroxide solution with a mass concentration of 3% for 5 minutes;

[0097] (2) Put the soaked P(CL-DLLA) stent into 0.40mol / L benzophenone (BP) solution and let it stand for 1h;

[0098](3) Take out the P(CL-DLLA) stent to dry, and place it under a 125W high-pressure mercury lamp for close-range irradiation for 15 minutes after drying;

[0099] (4) Add 4% ethanol solution of acrylic acid (volume ratio) to the P(CL-DLLA) stent and irradiate for 45 minutes under a UV lamp;

[0100] (5) After irradiation, take it out and wash it with a phosphate buffer with a pH of 7, soak overnight in a phosphate buffer with a pH of 7 at room temperature, and dry to obtain the acrylic ac...

[0105] The characterization of embodiment 2 RSV-AA-P (CL-DLLA) scaffold material

[0106] 1. Fourier Transform Infrared Spectroscopy (FTIR)

[0107] 1. Experimental method

[0108] The P (CL-DLLA), AA-P (CL-DLLA), and RSV-AA-P (CL-DLLA) in Example 1 are dried, and then an appropriate amount of samples are put on glass slides for measurement on the machine .

[0109] 2. Experimental results

[0110] The result is as figure 2 -A, figure 2 -1723cm in A-a -1 and 3439cm -1 There are two strong absorption peaks at , which are the C=O and O-H stretching vibration peaks in the bulk material P(CL-DLLA); figure 2 -1730cm in A-B -1 The absorption peak is reflected by the stretching vibration of C=O on the acrylic modified support, 3441cm -1 The absorption peak is the stretching vibration of O-H on the material; figure 2 -A-c in 1638cm-1 absorption peak is C=O stretching vibration on the material, 3305cm -1 The absorption peak is the stretching vibration of O-H, 1421cm -1 ...

Embodiment 3

[0128] Example 3 Biological effects of RSV-AA-P (CL-DLLA) scaffold material

[0129] 1. Analysis of RSV release

[0130] In order to further verify whether the RSV-AA-P (CL-DLLA) material synthesized in Example 1 has been successfully grafted with RSV, the synthetic material was used for liver seed cell-HSC cell culture, and the RSV level in the culture medium was analyzed by HPLC.

[0131] 1. Experimental method

[0132] HSC cells in the logarithmic growth phase were made into cell suspension, and different cell amounts were inoculated onto RSV-AA-P (CL-DLLA) materials, placed in 96-well culture plates and cultured for 7 days, collected at the same time every day Media in culture plates. Take the culture medium and add 0.8mL ethyl acetate, shake and extract for 15min, after standing for layers, take the upper organic phase and rotary evaporate to dryness at 40°C, and dilute the residue to 1mL with methanol. Detection was performed after filtration with a 0.45 μm filter mem...

Abstract

The invention discloses a scaffold material with the function of delaying cell aging and applications thereof. The scaffold material is a polylactic acid-caprolactone scaffold material grafted with resveratrol. The characterization and cell culture research on the scaffold material provided by the invention show that the modified scaffold material not only overcomes the defects that the hydrophobic scaffold body and too-small pores are beneficial to cell growth due to the addition of hydrophilic groups, but also can obviously delay cell aging. The material is simple and easy in modification method, has obvious effect, is used for nourishing liver seed cells, has the effects of delaying cell aging and improving seed cell activity, and lays a good foundation for liver tissue engineering preparation. The invention provides a new method for solving the problem of cell life in liver tissue engineering research.

Description

technical field [0001] The invention relates to the technical field of tissue engineering, more specifically, to a scaffold material capable of delaying cell aging and its application. Background technique [0002] my country is an area with a high incidence of liver diseases, and various liver diseases seriously endanger human health. Liver transplantation is currently the main way to treat acute and chronic liver failure and other serious liver diseases. However, limited by the source of liver donors, its clinical application faces great difficulties. As an ideal way to sustainably provide liver tissue, tissue engineering is expected to become an effective solution to the clinical dilemma of liver diseases. Liver tissue engineering is an emerging discipline that combines the principles of biology and engineering. Its core is to establish a three-dimensional space complex of cells and biomaterials, that is, living tissue with vitality, which is used to treat diseased tiss...
