Problems solved by technology

[0005] Chinese patent CN 108904471 A discloses a nano drug carrier Au / MnO 2 Its preparation method and application, the nano-drug carrier has a core-shell structure, with gold nanorods AuNRs as the core, mesoporous manganese dioxide MnO 2 As the shell, the drug carrier is used to carry anti-tumor drugs. Through NIR laser irradiation, AuNRs will have the phenomenon of surface plasmon resonance coupling, which has the ability of photothermal conversion and can dissociate the interaction between the carrier and the drug. The drug is released and stimulated by external energy for tumor targeting therapy; however, the Au nanorods in this patented technology have strong near-infrared absorption, which is not suitable as a smart reagent, so that the near-infrared absorption can be achieved from scratch, which is very difficult. It is difficult to rule out the interference of signals in normal tissues, which affects the imaging effect
In addition, the technology uses mesoporous MnO 2 Used to load drugs to achieve therapeutic effects on tumors, this patent does not use manganese dioxide as an activatable chemical kinetic intelligent reagent

Embodiment 1

[0043] A glutathione-triggered tumor diagnosis and treatment reagent, the preparation and testing process is as follows:

[0044] (1) Preparation of Au nanoparticles: 10mg HAuCl 4 Dissolve in 500mL deionized water and heat to boiling with stirring. Then 3 mL of sodium citrate solution (1 wt%) was added and boiling was continued for 30 minutes to obtain an Au seed solution. The Au seed solution was then cooled and transferred to a constant temperature water bath at 85 °C. Finally add 3 mL of sodium citrate (1 wt%) and 1 mL of HAuCl 4 (1wt%) solution in the seed solution, repeat this step once to four times every 15 minutes, and prepare a mixed solution A of gold nanospheres with a particle diameter of about 25 nm.

[0045] (2) Preparation of Au@MnO 2 Nanoparticles: Under vigorous stirring, 20 mL of KMnO 4 (10mM) solution was slowly added dropwise to 80mL mixed solution A (concentration of sodium citrate solution was 0.3mg / mL). Stir rapidly at room temperature for 1 hour a...

Embodiment 2

[0059] A glutathione-triggered tumor diagnosis and treatment reagent, the preparation and testing process is as follows:

[0060] (1) Preparation of Au nanoparticles: 5mL of chloroauric acid (1wt%) was added to 500mL of water, then 6.5mL of sodium citrate solution (1wt%) was added, and then boiled for about 30 minutes to prepare gold nanoparticles.

[0061] (2) Preparation of Au@MnO 2 Nanoparticles: Under magnetic stirring, add 42 mg KMnO to the gold nanoparticle solution (100 mL) and 4 . After reacting for 5 minutes under magnetic stirring, 50 mg of PAH dispersed in 1 mL of water was added dropwise to the solution, and the color changed from purple to brown. Then the precipitate was collected by centrifugation at 8000 rpm, and then washed and centrifuged with deionized water. Finally, pure Au@MnO was obtained 2 Aqueous solution of nanoparticles.

[0062] (3) Au@MnO 2 The nanoparticle solution (5mg / Kg) was injected into the tumor site and the subcutaneous side of the con...

[0066] A glutathione-triggered tumor diagnosis and treatment reagent, the preparation and testing process is as follows:

[0067] (1) Preparation of Au nanoparticles: 10 mL of chloroauric acid (1 wt%) was added to 500 mL of water, then 6.5 mL of sodium citrate solution (1 wt%) was added, and then boiled for about 30 minutes to prepare gold nanoparticles.

[0068] (2) Preparation of Au@MnO 2 Nanoparticles: Under magnetic stirring, add 21mg KMnO to the gold nanoparticle solution (100mL) and 4 . After reacting for 5 minutes under magnetic stirring, 100 mg of PAH dispersed in 1 mL of water was added dropwise to the solution, and the color changed from purple to brown. Abstract

The invention relates to a glutathione-triggered tumor diagnosis and treatment reagent. The tumor diagnosis and treatment reagent is a core-shell mechanism, with gold nano-spherical particles as the core and MnO2 as the shell. When Au(at)MnO2 with the core-shell structure contacts with glutathione, the MnO2 shell is reduced to Mn<2+> by endogenous GSH, and the Au core is also induced to agglomerate by glutathione due to the loss of shell protection. The generated Mn<2+> can be used for MRI sialography and chemical kinetic therapy, and the strong near-infrared absorption generated after Au agglomeration can be used for photoacoustic imaging and photothermal treatment. The reagent can be used as an intelligent diagnosis and treatment reagent triggered by a tumor microenvironment. By the useof the reagent, the signal interference of normal tissues during tumor imaging diagnosis can be effectively reduced and the damage to normal tissues during treatment can be alleviated. The invention provides a new material and a new idea for accurate diagnosis and efficient treatment of tumors, and the reagent of the invention has a broad application prospect.

Description

technical field [0001] The invention relates to the fields of nanomaterials and biomedicine, in particular to a glutathione-triggered tumor diagnosis and treatment reagent. Background technique [0002] Malignant tumors are still the main diseases that threaten human life and health. Due to the complexity of malignant tumors that are easy to be missed, misdiagnosed, and difficult to cure, traditional treatment methods such as surgery, radiotherapy, and chemotherapy can no longer meet the needs of early diagnosis and efficient treatment of tumors. The integration of diagnosis and treatment technology is an efficient therapy that combines diagnosis and treatment, and diagnosis guides treatment. Among the various diagnostic and therapeutic reagents currently developed, the diagnostic and therapeutic reagents for photoacoustic and nuclear magnetic resonance (MR) imaging-mediated combined photothermal and chemokinetic therapy have rarely been reported. Among them, photoacoustic...
