According to the method, through soaking and embedding culture, germination of the Huaiyu shan high-mountain miniature potatoes can be effectively promoted, the germination rate of the Huaiyu shan high-mountain miniature potatoes is greatly increased, and meanwhile germination and seedling formation of embedded beads of the Huaiyu shan high-mountain miniature potatoes are promoted through irrigation of the MS culture solution and alternate culture with day and night temperature difference; and therefore, the germination rate of the embedded beads of the Huaiyu shan high-mountain miniature potatoes is effectively increased, plants are promoted to be robust, sufficient nutrients can be provided, and follow-up culture and transplanting survival are facilitated.
