Pharmaceutical Composition, Preparation Method and Use Thereof

The present disclosure relates to the technical field of medicine, and in particular to a pharmaceutical composition, preparation method and use thereof. The pharmaceutical composition includes a first active ingredient, a second active ingredient, and a pharmaceutically acceptable carrier or excipient. The first active ingredient is a microbial agent, including one or more of Staphylococcus aureus, Bordetella pertussis, diphtheria toxoid, tetanus toxoid, Salmonella typhi, and Salmonella paratyphi. The second active ingredient includes polyinosinic acid, polycytidylic acid, and vitamin. The pharmaceutical composition of the present disclosure pertains to an artificial active immunotherapy for tumors. It can “stimulate” the entire immune system, making the therapy of using bacteria to activate the human immune system to kill cancer cells highly stable and reliable. This composition can significantly save and prolong the lives of cancer patients while exhibiting extremely high safety, minimal toxic side effects, and low production costs.

Immunogenic proteins from bordetella pertussis

Recent evidence accumulating over the last decade demonstrates that generation of CD4+ T cells is critical for sustained immunity against Bordetella pertussis. B. pertussis contains hundreds of antigens that are processed and presented on MHC Class II and recognized by CD4′T cells. The present disclosure relates to a vaccine comprising Bordetella pertussis antigen peptides to prevent infection of the Bordetella pertussis bacterium.

Freeze-dried live Bordetella vaccine

This invention provides a method for producing a freeze-dried vaccine containing attenuated Bordetella bacteria. [Solution] A method for producing a freeze-dried vaccine includes the steps of: collecting Bordetella bacteria from a culture; mixing the collected Bordetella bacteria with a freeze-drying buffer; freeze-drying the mixture of Bordetella bacteria and the freeze-drying buffer, wherein the holding time between the collection step and the freeze-drying step is less than 48 hours, and the freeze-drying step includes a pre-crystallization holding step, in which the mixture of Bordetella bacteria and the freeze-drying buffer is held for 0.5 to 10 hours at a temperature 0.1 to 10°C higher than the crystallization temperature of the mixture before further cooling; and collecting the freeze-dried Bordetella bacteria.
