Embodiment 1

[0018] Weigh 5g of oligomeric guluronic acid (weight-average molecular weight: 3.7kD) and dissolve it in 200ml of pure water, adjust pH=8 with 2mol / L sodium hydroxide solution, add 2mol dropwise under stirring in a water bath at 70°C / L chromium trichloride solution, keep using 2mol / L sodium hydroxide to maintain its pH at about 8 in the dropwise addition of chromium solution, stop adding sodium hydroxide and chromium chloride, and continue to keep warm for 2h. Take out the reaction solution, filter, add 3.5 times the volume of 95% ethanol, precipitate, let stand, collect the precipitate, dehydrate with absolute ethanol, and dry under reduced pressure at 45°C to obtain the chromium oligoguluronic acid complex, the measured chromium Content 4.1% (W / W, the same below).

Embodiment 2

[0020] Weigh 6g of oligomeric guluronic acid (weight-average molecular weight: 1.2kD) and dissolve it in 200ml of pure water, adjust the pH to 5 with 1mol / L sodium hydroxide aqueous solution, and add 2mol / L L chromium trichloride aqueous solution, and use 2mol / L sodium hydroxide to maintain its pH at about 5. When green flocculent precipitates appear in the solution, stop adding sodium hydroxide and chromium chloride, and continue to keep warm for 1h. Take out the reaction solution, filter, add 5 times the volume of 95% ethanol, precipitate, stand still, collect the precipitate, dehydrate with absolute ethanol, and dry under reduced pressure at 45°C to obtain the chromium complex of oligomeric guluronic acid. Content 12.4%.

Embodiment 3

[0022]Weigh 10g of oligomeric guluronic acid (weight average molecular weight: 9.7kD) and dissolve it in 500ml of pure water, adjust pH=10 with 2mol / L sodium hydroxide aqueous solution, add 2mol / L L chromium trichloride solution, and use 2mol / L sodium hydroxide to maintain its pH at about 10. When green flocculent precipitates appear in the solution, stop adding sodium hydroxide and chromium chloride, and continue to keep warm for 2 hours. Take out the reaction solution, filter, add 3 times the volume of 95% ethanol, precipitate, let it stand, collect the precipitate, dehydrate with absolute ethanol, and dry under reduced pressure at 45°C to obtain polyguluronic acid chromium complex, the measured chromium content 9.2%.

Abstract

The invention relates to an oligosaccharide metallic compound. The compound is characterized in that sugar residues of the compound consist of alpha-1, 4-L-guluronic acid; the weight average molecular weight of the compound is less than or equal to 12kD; and a molecular general formula of the compound is (C6H7O6)nCrm, wherein n is equal to 1 to 54, m is equal to 1 to 27 and the mass percentage of trivalent chromium is 0.1 to 12 percent. The preparation of the compound comprises the following steps: slowly adding chromic salt and dilute alkaline aqueous solutions in oligoguluronic acid aqueous solution under the conditions that pH is equal to 5 to 10 and temperature is between 40 and 80 DEG C; after stopping adding dilute alkaline and chromic salt aqueous solutions, carrying out heat-insulation reaction for 1 to 3h; carrying out filtration and precipitating the filtrate by ethanol with the volume 3 to 5 times that of the filtrate; carrying out static, precipitation and centrifugation; and collecting and washing the precipitate by using anhydrous ethanol to obtain the compound after drying. The oligosaccharide metallic compound has the advantages that because the raw materials are derived from marine natural products, the compound has abundant resources, easy industrialization, high safety, unique structure and ideal oral absorption and particularly integrates the advantages of marine oligosaccharide and trivalent chromium; and the compound not only has the functions of preventing insulin resistance and adjusting sugar and fat metabolism disturbance, but also has broad market and application prospects particularly in the prevention and treatment of diabetes II.

Description

technical field [0001] The invention relates to a sugar metal complex and its preparation method and application, in particular to an oligomeric α-L-guluronic acid chromium complex and its preparation method and its role in preventing and treating insulin resistance and improving glucose and lipid metabolism applications in disorders. Belongs to the field of biotechnology. Background technique [0002] Diabetes mellitus is a complex metabolic disorder affected by multiple factors, resulting from the interaction between environmental factors and genetic susceptibility, which can induce a series of complications, such as obesity, cardiovascular and cerebrovascular diseases, hypertension, Hyperlipidemia, nervous system lesions, renal failure and tumors, etc. Diabetes is divided into type I and type II, and more than 95% of diabetic patients belong to type 2 diabetes. One of the fundamental causes of diabetes is insulin resistance, and insulin resistance runs through the whole...
