Embodiment 1

[0042] Embodiment 1. Preparation of micro-detection chip

[0043] (1) Preparation of circular PMMA substrate

[0044] A stainless steel hole picker with an outer diameter of 16mm and an inner diameter of 13mm was placed on a laser turntable, and a circular substrate was placed on a PMMA substrate with a thickness of 4.8mm and 1.6mm respectively, and the substrate was placed in deionized water and anhydrous Ethanol, ultrasonic cleaning (250W) in particle-removing water for 20 minutes, and then dried in a blast oven.

[0045] (2) Preparation of upper porous substrate

[0046] The chip design graphics are drawn with CAD software, printed on the paper by laser, and the upper substrate is covered with paper, and the twisted brick with an outer diameter of 1.3mm is turned on the laser transfer table according to the drawing to form a porous substrate. The substrate was cleaned in deionized water, absolute ethanol, and deionized water by ultrasonic (250W) for 20 minutes, and then d...

Embodiment 2

[0049] Example 2. Preparation of functionalized magnetic nanoparticles

[0050] (1) Synthesis of superparamagnetic nanoparticles

[0051]Adopt co-precipitation method to synthesize superparamagnetic nanoparticles, specific method is as follows, 324.4mg ferric chloride hexahydrate, 119.3mg ferrous chloride tetrahydrate and 4.5g dextran (Dextran) (molecular weight=10T) are dissolved in 10mL to In deionized water, cool to 2-4°C, add 450 μL 2-4°C ammonia water (25-28%) under strong mechanical stirring and nitrogen protection, and heat the reaction mixture uniformly to 75-85°C within one hour and maintain at this temperature 75 minutes. After the reaction solution was naturally cooled to room temperature, impurities and excess dextran were removed by dialysis (molecular weight cut-off = 300KD), and finally the solution was concentrated by ultrafiltration (300KD).

[0052] (2) Surface modification of superparamagnetic nanoparticles

[0053] 5mL of the superparamagnetic nanopartic...

Embodiment 3

[0055] Example 3. Detection of Transverse Relaxation Time

[0056] PBS solutions of different concentrations of bovine serum albumin (BSA) and avidin (Avidin) were mixed with bovine serum albumin antibody-modified magnetic nanoparticles and biotinylated magnetic nanoparticles and kept at a constant temperature of 38°C for 90 minutes in a water bath Finally, T2 was measured in a low-field magnetic resonance analyzer with a magnetic field strength of 0.47 Tesla, and the pulse sequence used was Carr-Purcell-Meiboom-Gill (CPMG) sequence.

Abstract

The invention belongs to the field of magnetic resonance imaging and relates to a magnetic relaxation switch-based detection method. According to the method, based on the change of transverse relaxation time T2 of magnetic nanoparticles in dispersion and aggregation states and caused gray scale change in a T2 weight image, specific target molecules are detected through functionalization modification on the surfaces of the magnetic nanoparticles. The method comprises the following steps of: mixing samples to be detected and corresponding functionalized magnetic nanoparticles; placing the mixture on a minitype detection chip; and detecting in a low field magnetic resonance analyzer. Since a minitype multi-sample detection chip with small size is used in the method, required sample quantity is small and multiple samples can be detected in parallel quickly on the low field magnetic resonance analyzer. According to the method, the problem that conventional imaging equipment has high price and large size, is difficult to operate and not suitable for routine lab application and the problem that the low field magnetic resonance analyzer has a small sample chamber and is not suitable for multi-sample parallel detection can be solved.

Description

technical field [0001] The invention belongs to the field of magnetic resonance imaging, and relates to a detection method based on a magnetic relaxation switch. In particular, it relates to a miniature detection method based on a magnetic relaxation switch suitable for application in a low-field nuclear magnetic resonance analyzer. Background technique [0002] Magnetic relaxation switches (Magnetic Relaxation Switches) are a kind of biosensor based on magnetic particles. Functional modification of the surface of magnetic particles can detect biologically active molecules such as oligonucleotides, deoxyribonucleic acid, proteins, enzymes, viruses, bacteria, and antibodies. Compared with traditional optical detection methods such as ultraviolet-visible spectroscopy and fluorescence spectroscopy, this detection method has very high sensitivity and specificity, and its advantage lies in the change of the relaxation rate (1 / T2) of the surrounding water protons after the magnet...
