Problems solved by technology

[0008] At present, collagen membranes mainly represented by Bio-Gide are widely used in foreign countries. Due to their high price, they are difficult to use in large quantities in China. However, the domestically produced membranes for guiding tissue regeneration have complex processes, high prices, and are not conducive to cell growth and proliferation. Defects in tissue repair and unstable osteogenesis
In addition, the vast majority of GTR membranes at home and abroad have no antibacterial effect

Embodiment 1

[0083] Preparation of spinning solution: Dissolve PLGA in a mixed solvent of chloroform and N,N-dimethylformamide (ratio 8:2-5:5) to make a 15% (w / v) PLGA solution, add Wool keratin, its concentration in the solution is 1.5% (w / v), fully stirred, then add ornidazole, its concentration in the solution is 3% (w / v), fully stirred and ultrasonic vibration for 60min, as spinning liquid for use.

[0084] The composite membrane was prepared by electrospinning technology. The specific process was to put the spinning solution into the syringe, grind the tip of a No. 7 metal needle and connect it to the syringe, place the syringe on the screw driver, and at the same time make the metal needle of the syringe contact with the syringe. High voltage power supply is connected. Turn on the power supply of the screw driver, the syringe containing the spinning solution is pushed by the screw driver and the flow is controlled, the spinning solution is supplied to the spinneret (needle) and then...

Embodiment 2

[0086] Preparation of spinning solution: Dissolve PLGA in a mixed solvent of chloroform and N,N-dimethylformamide (ratio 8:2-5:5) to make a 20% (w / v) PLGA solution, add Wool keratin, its concentration in the solution is 1.2% (w / v), fully stirred, then add ornidazole, its concentration in the solution is 5% (w / v), fully stirred and ultrasonically vibrated for 50min, as spinning liquid for use.

[0087] The composite membrane was prepared by electrospinning technology, and the specific process was the same as in Example 1.

Embodiment 3

[0089] Preparation of spinning solution: Dissolve PLGA in a mixed solvent of chloroform and N,N-dimethylformamide (ratio 8:2-5:5) to make a 16% (w / v) PLGA solution, add Wool keratin, its concentration in the solution is 1.0% (w / v), stir well, then add ornidazole, its concentration in the solution is 1% (w / v), stir well and ultrasonic vibration for 30min, as spinning liquid for use.

[0090] The composite membrane was prepared by electrospinning technology, and the specific process was the same as in Example 1.

Abstract

The invention relates to a guided tissue regeneration membrane with a bacteriostatic effect and a preparation method of the membrane. The guided tissue regeneration membrane is constructed by using antibacterial drug ornidazole, drug carrier poly(lactic-co-glycolic acid) and film-forming material wool keratin. The method adopts a high-voltage electrostatic spinning method to compound the antibacterial drug ornidazole with the synthetic polymer compound PLGA and the natural ingredient wool keratin, thereby constructing the composite guided tissue regeneration membrane having excellent physicaland chemical properties, a slow release effect on the ornidazole, good cell compatibility and the bacteriostatic effect. The composite membrane can not only block migration of fibroblasts from surrounding tissue into a defect area, and make periodontal ligament cells preferentially migrated, attached, proliferated and differentiated on the tooth root surface, thereby inducing regeneration of periodontal tissue; the composite membrane can play a bacteriostatic role within 7 days after guided tissue regeneration, prevent the occurrence of postoperative infection caused by bacteria, and ensure the effect of tissue regeneration; and the guided tissue regeneration membrane provided by the invention has the simple preparation method, mild film formation conditions, and good cell compatibility and bacteriostatic performance, and meets the requirements of biological in-vivo application, and has good application prospects as a biological membrane with the bacteriostatic effect.

Description

technical field [0001] The invention relates to the technical field of biomedicine, in particular to a tissue regeneration guiding membrane with antibacterial effect and a preparation method thereof. Background technique [0002] Periodontal disease is a chronic, nonspecific, infectious disease that causes the destruction of tooth supporting tissues (gingiva, periodontal ligament, alveolar bone, and cementum), eventually leading to tooth loosening and loss. It is one of the most common oral diseases in humans and the leading cause of tooth loss in adults. Periodontal disease not only affects oral health, but also has a certain impact on overall health. [0003] The goal of periodontal disease treatment is to prevent the progression of the disease and to regenerate the lost periodontal tissue. Among them, the regeneration of periodontal tissue is the key to successful treatment. Traditional treatment methods such as scaling, root planing, and bone grafting can only restore...
