The preparation method comprises the following steps: putting acetaminophen and ibuprofen into a container filled with a methanol or acetonitrile solvent according to a molar ratio of 1:(1-2), conducting oscillating at a constant temperature of 22-28 DEG C for 4-10 days, taking out a mixed solution, and conducting filtering to obtain a white crystal, namely the acetaminophen-ibuprofen pharmaceutical co-crystal.
