A germ tube is an outgrowth produced by spores of spore-releasing fungi during germination. The germ tube differentiates, grows, and develops by mitosis to create somatic hyphae. A germ tube test is a diagnostic test in which a sample of fungal spores are suspended in animal serum and examined by microscopy for the detection of any germ tubes. It is particularly indicated for colonies of white or cream color on fungal culture, where a positive germ tube test is strongly indicative of Candida albicans.

Single spore isolation and purification method of verticillium dahliae

The invention relates to a single spore isolation and purification method of verticillium dahliae. The method mainly comprises the following steps: an agar medium cylinder is taken from a water agar plate by the use of a glass capillary with diameter being 1mm and is picked onto a sterilized glass slide with a sterilization needle; a verticillium dahliae strain is activated in a PDA solid medium and the activated strain is cultured in a Czapek-Dox liquid medium; spore concentration is counted under a microscope; and the spore concentration is adjusted to 102-106 cfu to prepare a spore suspension; 1 microlitre of the spore suspension is sucked up and dripped onto the surface of the water agar cylinder on the glass slide; the glass slide is moved into a culture dish for constant temperature incubation at 25 DEG C; the glass slide is detected under the microscope, and the cylinder containing a single conidiospore which has germinated germ tubes is transferred onto a PDA plate to be cultured at 25 DEG C so as to finish single spore isolation. The method is simple to operate, can be implemented by general operation staff and is suitable for mass separation. By the method, separation success rate is high. The method is also suitable for production of other fungi of microconidia.

The frass organic fertilizer is an extract of the Hermetia illucens frass in the methyl alcohol and is prepared by the method:the Hermetia illucens frass is smashed and then mixed with a methyl alcohol solution by vibration, the extract is obtained through freeze drying of a supernate obtained by ultrasonic separation, and an addition amount of the Hermetia illucens frass is 1g Hermetia illucens frass/9 mL methyl alcohol; and Stratiomyidae frass can be used for restraining germination of the weed broomrape, growth of germ tubes of the weed broomrape and germination and parasitism of Orobanche Cumana. According to the fertilizer, through joint effects of the Hermetia illucens frass and the methyl alcohol, germinationof the broomrape and growth of the broomrape germ tubes can be effectively restrained, the germination and parasitism of Orobanche Cumana can also be restrained, nutrient losses in a plant root systemcan be prevented, a plant death rate is reduced, and the yield and quality of plants are improved.

The invention relates to a method for separating and purifying the antibacterial peptide of an endophytic bacteria strain though high efficiency liquid chromatography, and the method comprises the following steps of: preparing the fermentation filter liquor of the strain by taking brevibacillus brevis B-011 as a basic strain, wherein the strain is collected in the China general microbiological culture collection center in April 12, 2013, and the collection number is CGMCC No.7466; adding acetonitrile; uniformly mixing, and standing; centrifugalizing; evaporating and concentrating a supernatant to obtain an antibacterial crude extract; and then carrying out secondary separation by adopting the high efficiency liquid chromatography to obtain an antibacterial peptide activity single-peak component. According to the invention, a bacteriostatic activity substance ( antibacterial peptide) contained in the fermentation filter liquor of the strain B-011 has inhibiting effect on the spore germination of alternaria solani and shows the teratogenic effect on germ tubes and hyphae. The method disclosed by the invention can be used for separating and purifying the bacteriostatic activity component contained in the strain fermentation filter liquor through the high efficiency liquid chromatography and identifying the substance and is conductive to defining the root cause of the strain on inhibiting the alternaria solani and developing the novel antibacterial drug.

Biological prevention and control technology for rice blast

The invention discloses a biological prevention and control technology for rice blast, and relates to the technical field of biological prevention and control of rice diseases and pests. The preparation method includes the steps of preparing a gradient diluent, preparing an NA solid culture medium, separating bacilli, preparing a PDA culture medium, preparing a spore solution, preparing extracts of murraya paniculata leaves and planting the rice. The bacilli separated according to the method can secrete antibiotic substances to prohibit growth of pathogenic bacteria and entice a plant defencesystem to resist invasion of pathogenic bacteria, and supplied testing bacterial strains can achieve an obvious prohibition effect on germination of mitogenetic spores, extension of germ tubes and formation of appressorium of the rice blast bacteria. The active substances in the bacterial strains have a great prohibition effect on the rice blast bacteria, and the prohibition rate to the mycelial growth of the rice blast bacteria reaches 80% or above. The extracts of the murraya paniculata leaves have a capability of resisting the pathogenic bacteria, insect antifeedant activity and a certain prohibition effect on the rice blast pathogenic bacteria, wherein the callus extracts have excellent bacteriostatic activity, and the bacteriostatic activity to the rice blast bacteria reaches 90% or above.
