Abstract

The invention relates to a high-efficient in-vitro propagation method of a 20-year-old schima superba big tree. The high-efficient in-vitro propagation method comprises the following steps of: taking a stem segment with axillary buds, which is grown on the 20-year-old schima superba big tree, as an explant, performing disinfection and sterilization, then inoculating into a starting culture medium, firstly placing under dark conditions for culturing for 7-12 days, further placing under an ordinary fluorescent lamp, performing illumination for 16h daily, culturing for 40 days under conditions that the temperature is 23-25 DEG C and the humidity is 50%-60%, and performing differentiation to grow into a test tube seedling; and shearing the grown test tube seedling, transferring into a propagation culture medium after high-temperature sterilization for propagation culture, then shearing the test tube seedling after propagation, then inoculating into a seedling strengthening culture medium after high-temperature sterilization for seedling strengthening culture, exterminating calli and part of the leaf blades after 32 days, retaining 3-4 blades, inoculating into a rooting culture medium after high-temperature sterilization for performing rooting culture for 12-15 days, taking out, cleaning, transplanting into a mixed matrix containing turf, yellow soil and perlite, then rooting and surviving after 25 days, wherein the volume ratio of the turf to the yellow soil to the perlite is 2: 1: 1, and the rooting rate is above 96%.
