Embodiment 1

[0038] 1) Preparation of thermosensitive chitosan / glycerophosphate hydrogel

[0039]Dissolve chitosan in 0.1mol / L dilute hydrochloric acid, stir and mix evenly at room temperature to make a chitosan solution (the mass percentage of chitosan in the solution is 1.5%), autoclave for 30 minutes, and keep at 4°C Store in the refrigerator for later use.

[0040] Sodium glycerophosphate was prepared into a solution with deionized water (the mass ratio of sodium glycerophosphate in the solution was 10%), sterilized with a filter, and stored in a refrigerator at 4°C for later use.

[0041] At 4°C, chitosan solution and sodium glycerophosphate solution were mixed in an equal volume ratio to form a mixed solution, and polyvinyl alcohol was added (the amount of polyvinyl alcohol accounted for 10% by mass of the mixed solution), and stirred to dissolve. Then add a certain amount of glutaraldehyde aqueous solution (the mass concentration of glutaraldehyde in the solution is 25%) for cross-...

[0047] 1) Preparation of thermosensitive chitosan / glycerophosphate hydrogel

[0048] Dissolve chitosan in 0.1mol / L dilute hydrochloric acid, stir and mix evenly at room temperature to make a chitosan solution (the mass percentage in the solution is 2.5%), sterilize with high pressure steam for 20 minutes, and store it in a refrigerator at 4°C for subsequent use .

[0049] Sodium glycerophosphate was prepared into a solution with deionized water (the mass ratio of sodium glycerophosphate in the solution was 12%), sterilized with a filter, and stored in a refrigerator at 4°C for later use.

[0050] At 4°C, chitosan solution and sodium glycerophosphate solution were mixed in an equal volume ratio to form a mixed solution, and polyvinyl alcohol was added (the amount of polyvinyl alcohol accounted for 20% by mass of the mixed solution), and stirred to dissolve. Abstract

The invention discloses a composite hydrogel material loaded with rosmarinic acid and a preparation method and application of the composite hydrogel material. The composite hydrogel material is prepared from liposome in which the rosmarinic acid is embedded and a temperature-sensitive type hydrogel, wherein the rosmarinic acid accounts for 0.1%-5% of the total mass of the liposome. The mixing mass / volume ratio of the rosmarinic acid-embedded liposome to the temperature-sensitive type hydrogel is 0.1 mg / ml-150 mg / ml. The rosmarinic acid-embedded liposome is prepared from lecithin and cholesterol, and the mass ratio of the lecithin to the cholesterol is (4 to 1) to (8 to 1). The liposome nanotechnology is adopted to embed the rosmarinic acid, the local concentration maintaining and deliveryof the rosmarinic acid can be more effectively realized, and a low-dose and high-efficiency therapeutic effect is achieved. Compared with pure rosemary acid, the antibacterial rate of the rosmarinic acid after being embedded in the liposome against staphylococcus aureus is significantly increased. The prepared composite hydrogel material is expected to be developed into a novel external-use material with a remarkable curative effect on acne, and good clinical application prospects are achieved.

Description

technical field [0001] The invention belongs to the technical field of medical composite hydrogel, and in particular relates to a composite hydrogel material loaded with rosmarinic acid, a preparation method and application thereof. Background technique [0002] Acne is a common chronic inflammatory disease of the pilosebaceous glands in adolescence, which belongs to the category of "lung wind acne" in traditional Chinese medicine. Clinically, it is characterized by blackheads, inflammatory papules, secondary abscesses or knots that occur on the face, chest and back, and is one of the main diseases that affect facial beauty. Traditional Chinese medicine believes that acne is mostly caused by blood heat in the lung meridians or damp heat in the spleen and stomach, flaming heat toxins or imbalance of Chong and Ren, strong liver, weak spleen and stomach, insufficient kidney yin, excess liver fire, or multiple factors combined to cause dampness, fat, turbidity, phlegm, blood sta...
