Abstract

The invention discloses a plant-derived exosome and a preparation method thereof, and application of the plant-derived exosome in preparation of drugs or scaffolds for animal tissue regeneration therapy. The preparation method comprises the following steps: (1) taking any part of a natural plant, performing a 2-(N-morpholino)ethanesulfonic acid buffer soaking and permeation treatment, then discarding a supernatant, collecting a treated wet sample, and performing refrigerated centrifugation to extract and obtain an apoplast solution, wherein the soaking and permeation treatment is specificallyas follows: vacuumizing is performed 2 to 5 times after 6 to 24 hours of soaking, a time for each vacuumizing process is independently 5 to 15 seconds, and an interval between every two adjacent vacuumizing processes is independently 10 seconds to 1 minute; and (2) performing ultrahigh-speed centrifugation on the apoplast solution to obtain the plant-derived exosome, wherein conditions of the ultrahigh-speed centrifugation are as follows: a centrifugal force is not less than 100000g, a centrifugation time is 1 to 7 hours, and a temperature is 0 to 4 DEG C.
