The invention provides an extraction method of medicinal and edible plant leaf protein and SOD (Super Oxide Dlsmutase), comprising the following steps of: pulping and extruding cleaned fresh leaves of mature silphium perfoliatum into juice; guiding the filtered juice to a heating tank for soaking, solidifying and sterilizing by heating to 75-90 DEG C, adding pure lactic acid accounting for 6%-8% of the mass of the juice when rapidly cooling the juice to 45 DEG C, and then precipitating 1.5 hours; filtering precipitated protein paste by using a filter press and then making a leaf protein humid cake; and drying in a spray drying method, carrying out vacuum packaging by using an aluminum plastic bag with a lining, and then storing in a clean storeroom at 0-4 DEG C. In the method, the whole extraction process is controlled within 2 hours, the activity of the extracted SOD is above 4000, and the method obviously shortens extraction time in the condition of ensuring the higher activity of a product, and saves a subsequent sterilizing link, thereby effectively saving industrial production cost.
