Abstract

The invention discloses an HPLC-MS / MS method for simultaneously measuring concentrations of three kinds of aminoglycoside antibiotics in plasma. According to the method, with Dabigatran as an interiorlabel, the protein in plasma is precipitated by using a methanol solution containing formic acid with the volume fraction of0.1%; drug components in the supernatant are separated by using high performance liquid chromatography; and then drug targeting detection is carried out by using a high-resolution mass spectrometry multi-reaction monitoring mode and quantification is performed, thereby realizing simultaneous analysis and measurement of the concentrations of three kinds of aminoglycoside antibiotics in plasma. The method has advantages of high speed, high targeting performance, rapidness,high throughput, high sensitivity, high specificity, high precision and accuracy, high stability, high extraction recovery rate, obvious matrix effect prevention, and dilution effect prevention. Themethod can be applied to the blood concentration monitoring of the conventional aminoglycoside antibiotics clinically, thereby guiding the rational clinical application of aminoglycoside antibiotics.
