Embodiment 1

[0026] 1. Obtaining a kind of bacillus:

[0027] Sampling time: June 2018.

[0028] Sampling location: The soil was collected from the saline-alkali land in Binhai New Area, Tianjin (38°46′N, 117°13′E).

[0029] Processing of samples: Gradient dilution of samples: Gradient dilution of samples with sterile physiological saline, select 10 -10 -10 -7 Experiment with serial dilutions.

[0030] Media used for screening:

[0031] Nutrient agar (NA) medium: peptone 10g, beef extract 3g, sodium chloride 5g, distilled water to 1L, adjust pH to 7.4, add agar 20g, sterilize at 121°C for 20min.

[0032] Potato dextrose agar (PDA) medium: 200g potatoes, 20g glucose, distilled water to 1L, natural pH, add 20g agar, sterilize at 115°C for 30min.

[0033] Landy liquid medium: glucose 20g, L-glutamic acid 5g, MgSO4 7H 2 O 0.5g, KH 2 PO 4 1g, KCl 0.2g, yeast extract 1g, L-phenylalanine 0.02g, sodium citrate 0.01g, ammonium sulfate 2g, FeSO 4 ·7H 2 O 5mg, MnSO 4 ·H 2 O 10mg, CuSO 4 ...

Embodiment 2

[0084] A fermentation method for Bacillus veles, comprising the steps of: adding landy liquid culture medium in proportion to a filling volume of 80ml / 250ml into a culture container, and adding the vesicles to a culture container at a ratio of 6% by inoculation volume Bacillus was picked into landy liquid medium, cultured at 30°C, 200rpm, on a constant temperature shaker for 48 hours, cooled to 4°C, centrifuged, and the precipitate was discarded. The supernatant was filtered through a 0.45 μm membrane to remove bacteria, and stored at 4°C for later use.

[0085] Carry out Plackket Burman test to bacterial strain XW0513, according to Plackket Burman test and the steepest climbing test result, design central combination (CCD) test, through above experiment, bacterial strain XW0513 can reach 12.68mm to Alternaria solani bacteriostatic zone width, is 2.33 times before optimization.

Embodiment 3

[0087] A fermentation method for Bacillus veles, comprising the steps of: adding landy liquid culture medium in proportion to a filling volume of 60ml / 250ml into a culture vessel, and adding the veles to a culture vessel at a ratio of 4% by inoculation volume Bacillus was picked into landy liquid medium, cultured at 28°C, 180rpm, on a constant temperature shaker for 50 hours, cooled to 4°C, centrifuged, and the precipitate was discarded. The supernatant was filtered through a 0.45 μm membrane to remove bacteria, and stored at 4°C for later use.

Abstract

The invention discloses bacillus velezensis XW0513 with a preservation number of CGMCC No.20474. The bacillus velezensis has an antifungal capability. The bacillus velezensis disclosed by the invention has an obvious inhibition effect on pathogenic fungi including botrytis cinerea, root rot fungi, fusarium oxysporum f.sp cucumerinum, fusarium moniliforme, pyricularia grisea, magnaporthe oryzae andaspergillus flavus, and is wide in antifungal spectrum. The bacillus velezensis has no antibacterial effect on pathogenic bacteria. It is shown that the strain XW0513 has a relatively good inhibitioneffect on various pathogenic fungi.

Description

technical field [0001] The invention belongs to the field of biological technology, and relates to a bacillus velei and a fermentation method and application thereof. Background technique [0002] Today, crop diseases are becoming more and more serious, in order to ensure the normal growth of crops and increase production, pesticides have become necessary for agricultural development. Due to the low price, variety and quick effect of chemical pesticides, chemical pesticides are still used in most areas. However, the long-term abuse of chemical pesticides has caused large-scale environmental pollution, endangering many beneficial organisms in the field, and has also seriously threatened human health. Fortunately, this phenomenon has attracted widespread attention all over the world. With the increasing awareness of people's health and environmental protection, the use of chemical pesticides is generally restricted or even eliminated on a global scale. However, the use of bio...
