The present application belongs to the technical field of detection reagents, and particularly relates to a
dermatophyte detection kit based on a recombinant LGBP-FITC fluorescent probe and application thereof. The present application obtains LGBP-Cys
protein by performing C-terminal
cysteine site-directed
mutation on LGBP
protein from
Procambarus clarkii; then, the LGBP-Cys
protein is directionally coupled with
maleimide-activated FITC under mild neutral conditions, so that the problem of
protein activity damage caused by traditional random
coupling is solved; finally, the
coupling product is placed in a composite stabilizing solution containing an anti-
quenching agent, so that the stability of the probe and the detection
signal-to-
noise ratio are significantly improved. The kit is simple to operate, and detection can be completed within 15 minutes. The kit has high sensitivity, high specificity and
visualization advantages for common clinical dermatophytes, and is suitable for rapid bedside diagnosis.
