Embodiment 1

[0036] Embodiment 1: 0.1 g chitosan and 0.029 g Ca(NO 3 ) 2 .4H 2 O was dissolved in 5 mL, 2% acetic acid to obtain a chitosan solution containing calcium ions. Add this solution into 100 mL soybean oil containing span-85, and stir well at room temperature. Subsequently, 0.027 g Na was added to the above system 2 HPO 4 .12H 2 Aqueous solution prepared by O (calculated according to the theoretical molar ratio of Ca / P in HA of 1.67). After stirring to make it evenly mixed, 1 mL of glutaraldehyde solution was added for cross-linking. After cross-linking for 4 h, 1M NaOH solution was slowly added dropwise to the emulsion system to adjust the pH value of the solution so that chitosan and HA could be deposited simultaneously. After continuous stirring at room temperature for 24 h, 20 mL of acetone was added and stirred for 30 min. The obtained product is centrifuged, washed repeatedly with acetone and water, and placed in the air at room temperature to finally obtain a compo...

Embodiment 2

[0037] Embodiment 2: 0.1 g chitosan and 0.059 g Ca(NO 3 ) 2 .4H 2 O was dissolved in 5 mL, 2% acetic acid to obtain a chitosan solution containing calcium ions. Add this solution into 100 mL soybean oil containing span-85, and stir well at room temperature. Subsequently, 0.053 g Na was added to the above system 2 HPO 4 .12H 2Aqueous solution prepared by O (calculated according to the theoretical molar ratio of Ca / P in HA of 1.67). Afterwards, the operating steps are the same as in Example 1. Finally, the composite microspheres with the mass ratio of chitosan and HA of 4 / 1 were obtained.

Embodiment 3

[0038] Embodiment 3: 0.1 g chitosan and 0.028 g anhydrous CaCl 2 Dissolve in 5 mL, 2% acetic acid to obtain a chitosan solution containing calcium ions. Add this solution into 150 mL soybean oil containing span-80, and stir well at room temperature. Subsequently, 0.020 g (NH 4 ) 2 HPO 4 Prepared aqueous solution (calculated according to the theoretical molar ratio of Ca / P in HA of 1.67). Afterwards, the operating steps are the same as in Example 1. Finally, the composite microspheres with the mass ratio of chitosan and HA of 4 / 1 were obtained.

Abstract

The invention relates to a method for preparing chitosan / hydroxyapatite composite microspheres, and belongs to the field of preparation technologies of biomedical materials. According to the method, chitosan, calcium nitrate and disodium hydrogen phosphate are taken as raw materials, and an in-situ synthesis technology is adopted for preparing the chitosan / hydroxyapatite (HA) composite microspheres. The method comprises the following steps: dropping a prepared calcium nitrate / chitosan solution into an oil phase containing Span 85 to form a water-in-oil type emulsion; adding a disodium hydrogen phosphate solution and uniformly stirring; dropping a glutaraldehyde water solution for cross-linking; finally regulating the pH value of a system through sodium hydroxide; performing dehydration, centrifugation, washing and drying to get a composite microsphere product. The chitosan / HA composite microspheres prepared by the method disclosed by the invention have the advantages of regular sphericity, narrow particle size distribution and high dispersivity, and can be used for effectively avoiding the problem of agglomeration of nano-HA particles during the preparation process of a traditional emulsification method, and be used as a medicine carrier or an injectable bone tissue repair material.

Description

technical field [0001] The invention relates to a preparation technology of biomedical materials, in particular to a method for preparing chitosan / hydroxyapatite composite microspheres. Background technique [0002] Composite materials are one of the most active fields in biomaterials research. So far, various forms of composite materials including porous scaffolds, membranes, fibers, coatings and microspheres have been successfully prepared and used in the repair of bone defects. Among them, the microsphere material has unique advantages such as small particle size, large specific surface area, and good fluidity. The injection-type artificial bone repair material made of microspheres has strong clinical operability, and is not limited by the range of bone defects when used, and is expected to change the traditional surgical bone grafting mode, with good safety and less trauma, so it has excellent performance It has more attractive application prospects than other forms of...
