Embodiment 1

[0027] Sampling the sediments of a mariculture area, the concentration of ammonia nitrogen in the sediments was detected to be 238mg / L, and the concentration of nitrate was 254mg / L. The immobilized bacterial agent package was used to test the ammonia nitrogen and Nitrate nitrogen inhibition experiment, including the following steps:

[0028] First, screen out efficient heterotrophic nitrifying and aerobic denitrifying bacteria: Weigh 10g of seabed sediment with a dry weight, add it to a 250mL Erlenmeyer flask filled with 100mL enrichment medium, and set the temperature at 150r / min at 28°C After several days of shaking culture, the supernatant was taken into the separation medium, and shaking culture was carried out under the above conditions for several days. Then, take 10 mL of the supernatant to a fresh separation medium, shake and culture at 150r / min, 28°C for several days, repeat the above steps for 3 times to increase the concentration of heterotrophic nitrifying and aero...

Embodiment 2

[0035] Sampling the sediments of a mariculture area, the concentration of ammonia nitrogen in the sediments was detected to be 46mg / L, and the concentration of nitrate was 39mg / L. Nitrate nitrogen removal experiment, including the following steps:

[0036] First, screen out efficient heterotrophic nitrifying and aerobic denitrifying bacteria: weigh 20g of seabed sediment with a dry weight, add it to a 250mL Erlenmeyer flask filled with 100mL enrichment medium, and set the temperature at 200r / min, 30°C After several days of shaking culture, the supernatant was taken into the separation medium, and shaking culture was carried out under the above conditions for several days. Then, take 10 mL of the supernatant to a fresh separation medium, shake and culture it at 200r / min, 30°C for several days, repeat the above steps for 3 times to increase the concentration of heterotrophic nitrifying and aerobic denitrifying bacteria, and finally Obtain heterotrophic nitrification aerobic den...

Abstract

The invention relates to a preparation method and application of an immobilized agent bag of an efficient denitrifier. The preparation method of the agent bag comprises the steps of screening out efficient heterotrophic nitrification and aerobic denitrification bacteria at first, and then carrying out salt-tolerant domestication on the efficient heterotrophic nitrification and aerobic denitrification bacteria to obtain salt-tolerant heterotrophic nitrification and aerobic denitrification bacteria; adsorbing the immobilized salt-tolerant heterotrophic nitrification and aerobic denitrification bacteria by using modified zeolite to obtain immobilized salt-tolerant heterotrophic nitrification and aerobic denitrification bacteria zeolite; and then flatly laying the obtain immobilized salt-tolerant heterotrophic nitrification and aerobic denitrification bacteria zeolite in a cloth bag to obtain the final product. The bacterial density of zeolite particles in the cloth bag is 1* 109-5* 109 CFU / mL. The prepared immobilized agent bag is widely used in shallow sea aquatic farms and aquaculture ponds, and efficient removal in a high-salinity environment is realized; and moreover, the agent bag can be reused, the cost is greatly reduced, the shortcoming that free microbial flora which are directly used easily flow along with tides of seawater, cannot accurately position bacteria and cannotbe in full contact with the bacteria is overcome effectively, threatening to the quality of aquaculture products due to introduction of alien microbes is also avoided, ecological safety of water of aseawater aquaculture region is protected, and the immobilized agent bag is expected to be widely used.

Description

technical field [0001] The invention belongs to water pollution control technology, and in particular relates to a preparation method and application of an immobilization of high-efficiency denitrification bacteria and a bacterial agent package. Background technique [0002] The rapid development of my country's mariculture industry has played an important role in meeting people's demand for seafood. However, during the production process, a large amount of pollutants produced by mariculture areas and farms directly enter the marine environment, and long-term accumulation leads to deterioration of water quality and imbalance of the marine ecosystem. Nitrogen source pollution in the process of aquaculture mainly comes from artificial baiting, animal manure, residual bait, and dead animals and plants, and exists in the form of nitrate nitrogen and ammonia nitrogen. Its existence can destroy the gill tissue of fish, induce gill rot, inhibit the oxygen-carrying capacity of the ...
