Embodiment 1

[0049] (1) Weigh 0.5g chitosan powder in a 100mL three-necked bottle, add 30mL mass fraction of 5% acetic acid solution, stir at room temperature, after the chitosan is fully dissolved, add 30mL liquid paraffin, and stir for 10 minutes;

[0050] (2) Warm up the product of step (1) to 50°C, add 2-3 drops of emulsifier Span80 dropwise, and emulsify for 10 minutes to form fine chitosan droplets;

[0051] (3) Add 1.0 mL of formaldehyde solution to the obtained product of step (2), and stir and react for 1.5 hours;

[0052] (4) Warm up the product of step (3) to 70°C, add dropwise 5-6 drops of NaOH solution with a mass fraction of 10%, keep the pH of the solution alkaline, and slowly add 2.0mL of epoxy chloride Propane, reacted for 5 hours;

[0053] (5) The gain of step (4) is filtered, washed with distilled water, then washed with sherwood oil, absolute ethanol, and water successively until the pH value of the washing liquid reaches neutrality, and chitosan microspheres (NCTS) are ...

Embodiment 2

[0097] (1) Accurately weigh 10 mg of modified chitosan microsphere carrier (NCTS-EDA), and separate after swelling with 10 mL of phosphate buffer solution (PBS, pH=7) for 24 hours;

Abstract

The invention discloses a magnetic chitosan microsphere for enzyme immobilization as well as a preparation method and an application of the magnetic chitosan microsphere. The preparation method comprises the following steps: adding acetic acid solution with weight percentage being 5% into 0.5g of chitosan powder, after the chitosan powder is completely dissolved, adding liquid paraffin into the mixed solution, stirring for 10min, heating to 50 DEG C, dropwise adding emulsifier Span 80, and emulsifying for 10min; adding 0.5-2.0 mL of formaldehyde solution into the obtained emulsion, and stirring for reaction for 1.5h; heating to 70 DEG C, dropwise adding with NaOH solution with the weight percentage being 10%, then slowly dropwise adding with 1.0-2.5 mL of epoxy chloropropane, and reacting for 5h; filtering, washing by using distilled water, petroleum ether, absolute ethyl alcohol and water, thus obtaining a chitosan microsphere (NCTS); adding 50mL of N,N-dimethylformamide (DMF) into 50mg of NCTS, and soaking overnight; and adding 250mg of ethylenediamine, stirring for reaction for 12h at 90 DEG C under the protection of nitrogen, carrying out heat treatment for 9h by adopting 1mol / L hydrochloric acid, carrying out alkali wash, acid pickling and water washing, and drying to constant weight, thus obtaining the modified chitosan microsphere (NCTS-EDA). The invention provides a good carrier for immobilized enzyme.

Description

technical field [0001] The invention relates to the field of chitosan microsphere preparation and application research, in particular to a modified chitosan microsphere for enzyme immobilization, a preparation method and application thereof. Background technique [0002] Cellulose is the most widely distributed, most abundant, and renewable natural organic substance, which can continuously provide energy for human beings. However, in nature, most of the cellulose will be decomposed and transformed by various microorganisms, and only about 11% of the cellulose raw materials are used in crop products, feed, pharmaceuticals, textiles, papermaking and construction, which not only causes a huge waste of resources , and pollute the environment, the harm is quite large. [0003] Enzymes are extremely sensitive to environmental conditions and are unstable to acids, alkalis, heat, metal ions, and some organic solvents. Generally, enzymes act on substrates in a water-soluble state, ...
