The application belongs to the technical field of reversing
liver fibrosis, and particularly relates to self-assembled nanoparticles for reversing
liver fibrosis and a preparation method and application thereof. The nanoparticles comprise N-acetylmannosamine and 2-(2-propylpyridine-4-yl)-4-(p-tolyl)
thiazole. The preparation method comprises the following steps: S1, self-
assembly reaction: under stirring, an organic phase solution of 2-(2-propylpyridine-4-yl)-4-(p-tolyl)
thiazole is added into an aqueous phase solution of N-acetylmannosamine, the mixed
emulsion is continuously stirred, and then ultrasonic treatment is performed on the mixed
emulsion to complete self-
assembly; and S2, purification: the reaction solution after S1 is allowed to stand,
centrifugation is performed to discard the supernatant, and then a washing solution is added into the centrifugal tube to resuspend the
centrifugation, and the purified self-assembled nanoparticles are obtained after
centrifugation. The application forms a stable
nanostructure through intermolecular self-
assembly of ManNAC and Fatostatin, realizes efficient enrichment and accurate release at a fibrotic
lesion in the liver, and reverses the
liver fibrosis process through synergistic regulation of a
lipid metabolism axis.
