The invention discloses an
agrobacterium rhizogenes mediated transformation method for transgenic hairy roots of complete leaves of cinnamon. The stable and repeatable
agrobacterium rhizogenes-mediated hairy root transformation
system is established for the first time aiming at the
cinnamomum cassia which is a
cinnamomum camphora
woody plant with difficulty in genetic transformation, and the technical difficulty in
gene function research is solved. It is found that in cinnamon, only on the premise that the overall integrity of leaves is kept, the stable
agrobacterium rhizogenes infection and hairy root induction can be achieved by slightly
scratching the leaves perpendicular to leaf veins in cooperation with an agrobacterium infection solution with a certain concentration and vacuumizing treatment, and the hairy root
induction effect is achieved. And a stable material is provided for subsequent hairy root induction of a complete transgenic regeneration
plant. The agrobacterium rhizogenes-mediated hairy root transformation method provided by the invention can efficiently induce plants to generate transgenic roots, lays a foundation for mining good cinnamon genes in the future, and provides a convenient and rapid way for researching biological functions of related genes in the roots.
