The method comprises the steps that firstly, after light supplementation treatment is conducted on stocks, sampling is conducted, and the antioxidant capacity of the stocks is analyzed; secondly, after grafting culture is conducted on the rootstock after light supplementing treatment, optimal rootstock light supplementing treatment is determined, and meanwhile the grafting survival rate of the rootstock after grafting is measured; thirdly, performing grouped light supplementing treatment on the watermelon grafted survival seedlings, performing destructive sampling every 7 days after light supplementing is started, and recording sampling results; and finally, obtaining growth characteristic indexes of the watermelon grafted seedlings according to a sampling result, so that the optimal light supplementing intensity of the watermelon grafted seedlings is obtained through the growth characteristic indexes. According to the optimal light supplementing intensity, the growth of scions and stocks is remarkably promoted, the disease index is reduced, the growth of tendrils is inhibited, and the net photosynthetic rate, the total chlorophyll amount and the activity of antioxidant enzymes such as SOD and CAT are remarkably improved.
