Problems solved by technology

While Paenibacillus nitrogen-fixing has nitrogen-fixing effect, it can also produce some substances that promote plant growth and some antibacterial substances, which can effectively improve the soil microenvironment, promote plant growth and enhance the ability of plants to resist diseases. Many strains in this genus are known as It has been widely studied as a growth-promoting bacteria in plant rhizosphere, but all of them are limited to large plants. At present, there is no example of Paenibacillus nitrogen-fixing as a growth-promoting bacteria in microalgae culture.

Embodiment 1

[0025] Example 1 Determination of Pig Raising Wastewater Components and Preparation of Diluted Pig Raising Wastewater

[0026] Pig raising wastewater in this example comes from Hongyun Farm in Cuicun, Changping, Beijing. The determination method of the total nitrogen (total nitrogen, TN) of pig raising wastewater is alkaline potassium persulfate digestion ultraviolet spectrophotometry, according to the national environmental protection standard HJ 636-2012 of the People's Republic of China; and the total phosphorus (total phosphor, TP) The determination method is ammonium molybdate spectrophotometry, according to the national standard GB118WLY93-93 of the People's Republic of China. Ammonia nitrogen (NH 3 -N) and COD are determined based on Lianhua Water Quality Rapid Testing Instrument 5B-6C. Determination of TN and NH in pig wastewater by the above method 3 -N, TP and COD components, the measurement results are listed in Table 1:

Embodiment 2

[0030] Embodiment 2 improves the method for the biomass of chlorella in pig raising wastewater

[0031] 1. Isolation, purification and cultivation of chlorella and autogenous nitrogen-fixing bacteria

[0032] (1) Isolation, purification and cultivation of Chlorella: solid plate TAP medium: agar powder 1.5%; pick the single clone of Chlorella sp.34-2 on the plate and inoculate it on the plate for preservation and purification by streaking method Algae were subcultured once a week.

[0033] Pick a single clone on the TAP plate of Chlorella sp.34-2 and inoculate it in 20ml of TAP liquid medium, then inoculate 1ml of the culture solution into 100ml of TAP liquid medium, and place it in a light incubator at 25°C. Set up culture.

[0034] (2) Isolation, purification and cultivation of Paenibacillus 18WLY: solid LB medium: agar powder 1.5%; single clone picked from Paenibacillus sp.18WLY was inoculated on the plate by streaking method to preserve and purify the strain, every three ...

Abstract

The invention provides a method for improving the biomass of chlorella in swine wastewater, belonging to the field of microbe application. Chlorella assigned with the accession number of CGMCC NO.9225 and paenibacillus elgii assigned with the accession number of CGMCC NO.7724 are co-cultured in the swine wastewater, the result shows that paenibacillus elgii can obviously improve the biomass of chlorella in the swine wastewater, and the promotion effect achieves 1.6 times. With the adoption of the method, the treatment problem of the swine wastewater is solved, and the environment pollution is reduced; due to the addition of paenibacillus elgii, the water body microenvironment in the swine wastewater is improved, the biomass of chlorella is obviously improved, further, the production cost of microalgae biodiesel is reduced, thus good economic benefit is obtained, and therefore, the method has a good market application prospect.

Description

technical field [0001] The invention belongs to the field of microorganism utilization and bio-oil preparation, and in particular relates to a method for increasing the biomass of chlorella in pig-raising wastewater. Background technique [0002] The oil-rich microalgae industry has become one of the most promising biodiesel industries and has attracted much attention. Compared with other energy crops, microalgae have many advantages, such as low input and high yield, no competition with farmers for land and fertilizer, can use waste gas, waste water, and salt water to grow, and do not require herbicides or pesticides. Microalgae cell residues can be used as agricultural fertilizer or animal feed or continue to produce biogas or ethanol, etc. [0003] At present, animal excrement pollution is becoming more and more serious, which not only leads to eutrophication of water bodies, but also causes harm to agricultural production, and also has a negative impact on people's livi...
