Problems solved by technology

For the prevention and control of root rot of Salvia miltiorrhiza, currently chemical pesticides are mainly used for control, but the control effect is very small; and the use of beneficial microbial agents to control fungal diseases of Salvia miltiorrhiza is the best choice for solving soil-borne diseases of Chinese medicinal materials

Embodiment 1

[0046] 1. Isolation of strain DS-R5

[0047] Take the healthy plants of Salvia miltiorrhiza that are growing well, clean them with running water, and disinfect the surface of the material according to the following procedures: soak in 75% ethanol for 1 min, rinse with sterile water for 3 times; soak in 0.1% mercuric chloride for 3 min, and rinse with sterile water for 3 times. Grind the treated Danshen roots, stems, leaves and other tissues with a mortar in a sterile operating bench, dilute them 10 times with sterile water, spread them on NA medium, and incubate at 37°C for 3 days. When visible colonies grow on the medium, pick the colonies in time, and obtain pure cultures by streaking multiple times.

[0048] Fusarium solani was used as the target pathogen to screen the endophytic antagonistic bacteria in Salvia miltiorrhiza. The endophytic bacteria isolated from Salvia miltiorrhiza were tested for their antagonism against pathogenic fungi using the two-point plate confront...

Embodiment 2

[0060] Embodiment 2: Determination of bacterial strain DS-R5 to common plant pathogenic fungus antibacterial spectrum

[0061] The endophytic bacterium DS-R5 isolated from Salvia miltiorrhiza was tested for its antagonism against pathogenic fungi by two-point plate confrontation method. Take Fusarium solani, Rhizoctonia solani, Alternaria, Fusarium graminearum, Fusarium oxysporum, Fusarium pseudograminea, Anthracnose bacteria, Vitis vinifera respectively cultured for 7 days, and beat them with a 5mm diameter Punch a hole with a holer, inoculate a 5mm bacterial cake in the center of the PDA plate, and inoculate the isolated endophytic bacteria of Salvia miltiorrhiza at a distance of 2.5cm from the bacterial cake. Use the fungal plate without antagonistic bacteria as a control, and place it in a 30°C incubator at a constant temperature to cultivate. When the pathogenic bacteria colonies of the control group covered the bottom of the dish, the width of the inhibition zone of the...

Embodiment 3

[0065] Embodiment 3: Bacterial strain DS-R5 is to common pathogenic bacteria antibacterial spectrum determination

[0066] Take a loop of Staphylococcus aureus, Escherichia coli, Candida albicans, Septicemia melanothorax, Candida albicans, and bacterial poplar canker into LB medium for overnight culture, and take 20 μL of the above-mentioned bacteria in the logarithmic phase Add 6 kinds of pathogenic bacteria to be tested into 20mL uncoagulated LB medium, mix well and pour into a sterile petri dish with a diameter of 7.5cm. After coagulation, inoculate strain DS-R5 with a sterilized toothpick, and observe the antagonistic effect of strain DS-R5 on the tested pathogenic bacteria after culturing at 37°C for 24 hours. Test result shows, bacterial strain DS-R5 all has inhibitory effect in varying degrees to 6 kinds of testing pathogenic bacteria (see Figure 4 ).

Abstract

The invention discloses paenibacillus polymyxa DS-R5 and preparation and application of a fungicide of the paenibacillus polymyxa DS-R5. The paenibacillus polymyxa DS-R5 is preserved in China Center for Type Culture Collection on January 4, 2021, and the biological preservation number is CCTCC M 2021005. The paenibacillus polymyxa DS-R5 has an inhibiting effect on various pathogenic bacteria, and particularly has a very strong antagonistic effect on fusarium solani, rhizoctonia solani and the like which cause root rot. The paenibacillus polymyxa DS-R5 can increase soil nutrient elements, further can improve salvia miltiorrhiza rhizosphere microbial community structure research, inhibits the number of pathogenic bacteria in soil so as to interfere infection of the pathogenic bacteria to salvia miltiorrhiza roots, and has dual effects of preventing diseases and promoting growth. Meanwhile, a microbial fertilizer taking the paenibacillus polymyxa DS-R5 as an active component is low in cost, high in fertilizer efficiency and free of pollution, and is an optimal substitute of chemical fertilizers and pesticides frequently used in prevention and treatment of root rot of salvia miltiorrhiza; and guiding significance is achieved for developing new traditional Chinese medicine salvia miltiorrhiza resources, reducing fungal diseases of salvia miltiorrhiza and improving the yield of salvia miltiorrhiza.

Description

technical field [0001] The invention relates to the technical field of agricultural microorganisms, in particular to the preparation and application of a strain of Paenibacillus polymyxa and its bacterial agent. Background technique [0002] Salvia miltiorrhiza (Salvia miltiorrhiza) is a perennial herbal medicinal plant. Its roots are used as medicine. It has the functions of dispelling blood stasis and relieving pain, promoting blood circulation and regulating menstruation, nourishing the heart and eliminating troubles. It is widely used in the treatment of cardiovascular and cerebrovascular diseases. Salvia miltiorrhiza is one of the medicinal materials with a large cultivation area. Under artificial cultivation conditions, due to the poor diversity and low richness of biological communities in the planting ecological environment, and the prominent dominant populations of diseases and insect pests, serious diseases and insect pests often occur, affecting the growth and deve...
