Problems solved by technology

However, DBM still has the following disadvantages as a bone graft substitute: 1. There are differences in osteoinductive and osteoconductive properties when processed in batches; 2. Although the antigenicity of DBM is weak, it will show severe immune rejection in a small nu...

Embodiment 1

[0026] Embodiment 1: prepare rabbit nanometer DBM

[0027] 1. The rabbit DBM bone block was prepared by the modified Urist method, and the steps were as follows:

[0028] 1) Materials: 1 New Zealand rabbit (provided by the Animal Experiment Center of the Second Military Medical University, the same below), 100g of fresh rabbit bone was routinely taken, the periosteum and all soft tissues were removed and split into bone pieces, the bone marrow was scraped off and purified with aseptic Rinse repeatedly with water, dehydrate in absolute ethanol for 2 hours, degrease with ether for 1 hour, and dry overnight in a ventilated place, then freeze-dry the bone in a -80°C refrigerator for later use;

[0029] 2) Preparation of DBM bone blocks:

[0030] (1) Degreasing:

[0031] Soak the above bones in 100ml of absolute ethanol for 2 hours in order to remove the ethanol; add 100ml of ether to soak for 12 hours and remove the ether; rinse with distilled water for 3 times to remove the eth...

Embodiment 2

[0040] Embodiment 2: preparation mouse nanometer DBM

[0041]20 mice (provided by the Animal Experiment Center of Second Military Medical University, the same below), male or female, weighing 25-30 g. 50 g of bone was routinely taken, and the modified Urist method was used to prepare allogeneic DBM bone blocks and make nano-DBM. The method was the same as in Example 1, and 12 g of nano-DBM was obtained.

Abstract

The invention relates to the technical field of biomaterial, and relates to a preparation method for a nanometer allograft decalcified bone matrix material DBM for bone graft. The preparation method comprises the following steps: 1) making a fresh allograft bone into DBM bone pieces conventionally by adopting an improved Urist method; 2) placing the DBM bone pieces into a liquid nitrogen refrigerating grinder to pre-grind, so as to obtain DBM powder; and 3) preparing the nanometer DMB, that is, placing the DBM powder in an ultrathin powder grinder, adding proper distilled water, grinding the DBM powder completely at a temperature of between 17 and 25 DEG C, treating the obtained homogenate after grinding by high-speed centrifugation, removing supernatant, then obtaining the nanometer DBM, and finally disinfecting the nanometer DBM for standby after molding and drying. The preparation method is convenient and uses the allograft bone as a raw material, so the material is easy to obtain; because the DBM prepared by the method is of a nanometer structure, the nanometer DBM can be used as a bone graft substitute, has good biocompatibility, can be used as a favorable carrier for exogenous growth factors such as bone morphogenetic protein and the like for promoting osteanagenesis, and has favorable capability for promoting osteanagenesis.

Description

technical field [0001] The invention relates to the technical field of biomaterials, and relates to a method for preparing a nano-DBM of an allogeneic decalcified bone matrix material for bone transplantation. Background technique [0002] Bone transplantation is the most common tissue transplantation after blood transfusion in clinical practice. The repair of bone defects caused by various reasons is a difficult problem for orthopedic surgeons to face. In addition, with the deepening of the understanding of diseases and the development of surgical techniques, the demand for bone transplantation in spine surgery, neurosurgery, etc. is also increasing. [0003] In the past 10 years, the research on bone graft substitutes has been one of the hot spots in the field of spinal surgery. Currently used bone graft substitute materials mainly include autologous bone, allogeneic bone and synthetic materials. An ideal bone graft should have the following three basic characteristics: ...
