Problems solved by technology

The paddy and dry rotation requires a long rotation period and no other hosts nearby to achieve good results, so it is difficult to implement
After the soil is sterilized, a soil biological vacuum is formed. Once the soil is infected by pathogenic bacteria, it will lead to a large outbreak of pathogenic bacteria and cause more serious consequences, and it is not feasible to operate in the field
Cultivating grafted seedlings with disease-resistant wood has obvious effects on the prevention and control of soil-borne diseases, but it can only be achieved with good wood and suitable grafting techniques, and large-scale applications also have certain limitations.

Embodiment 1

[0013] Embodiment 1: the acquisition of Pseudomonas fluorescens (Pseudomonas fluorescens) ②-16' bacterial strain

[0014] The invention provides a bacterial strain for controlling tomato bacterial wilt by using pathogenic bacteria and antagonistic microorganisms, specifically Pseudomonas fluorescens ②-16'. In 2008, experimenters collected soil samples from continuous cropping vegetable fields in different areas such as Jinhua, Taizhou, and Lishui, and used the bacterial culture medium to isolate more than 300 strains of various types of bacteria, and then purified the strains. The strain ②-16' antagonizing R. solanacearum was screened out by cultivating on slant and shaking flask under the culture condition of 28-30 ℃ and inoculating on various seedling substrates.

[0038] Preparation of beef extract peptone agar medium: same as 1 in Example 1.

[0039] 1.2 Liquid medium

[0040] Bacterial liquid shake flask medium: beef extract peptone medium without adding agar.

[0041] Liquid shake flask culture medium for R. solanacearum of tomato: the culture medium for R. solanacearum does not need to add agar.

[0042] 2. Test steps

[0043] 2.1 Culture medium preparation

[0044] Prepare Ralstonia solanacearum medium (solid, liquid) to prepare the slant of the pathogen Larsia solanacearum; prepare beef extract peptone agar ...

Embodiment 3

[0084] Embodiment 3: the preparation of the antagonistic agent of bacterial strain ②-16'

[0085] The microbial bacterial agent capable of antagonizing tomato bacterial wilt described in the present invention is formed by culturing and propagating Pseudomonas fluorescens ②-16' strain.

[0086] The specific operation process is as follows: Pseudomonas fluorescens ②-16' is based on beef extract medium, adding a small amount of inorganic salts such as calcium, magnesium, phosphorus, etc. for liquid shake flask fermentation, cultured at 25-35°C, and the rotation speed is 120-220rpm. Make the liquid parent species for bacterial classification inoculation liquid fermenter with this, liquid fermenter raw material is bran, vegetable cake and bean cake powder (ratio 1%~5%: 0~3%: 0~3%: 0~3% ) and a small amount of inorganic salts, etc., the amount of water added is 99-86%. After autoclaving, when the temperature of the liquid in the fermenter is cooled to below 35°C, inoculate the liqui...

Abstract

The invention provides a strain namely a pseudomonas fluorescens strain (2)-16' for controlling tomato bacterial wilt by using pathogenic bacteria antagonistic microbes, wherein the preservation serial number of the strain is CGMCC NO.3330. The invention also provides a microbial inoculum prepared from the pseudomonas fluorescens train (2)-16'. The invention further provides use of the pseudomonas fluorescens strain (2)-16' for controlling the tomato bacterial wilt, particularly as a seedling culture medium for controlling the tomato bacterial wilt. The pseudomonas fluorescens strain (2)-16' is directionally separated and selectively cultured, is cultured for propagation first, and then is prepared into a product of the microbial inoculum; mushroom residues after culturing edible fungi are fermented and thoroughly decomposed and then are used for preparing the seedling culture medium together with other base materials in certain proportion; and a raw anti-microbial agent is organically combined with the seedling culture medium directly. The pseudomonas fluorescens strain (2)-16' has a large number of advantages of abundant available resources, low cost and capacity of avoiding secondary pollutions, and provides an effective way for solving a soil-borne disease namely the tomato bacterial wilt.

Description

technical field [0001] The invention belongs to the field of biological technology, and in particular relates to a tomato bacterial wilt antagonistic strain and the field of biological control of tomato bacterial wilt by a seedling-raising substrate prepared by using the bacterial agent of the bacterial strain. Background technique [0002] Mushroom dregs are mainly discarded mushroom tubes after cultivation such as shiitake mushrooms and enoki mushrooms. The main components are wood dregs and cottonseed shells, which are mainly lignin and have low nutrient content. After being decomposed by edible fungi, the mushroom dregs are slightly acidic and soft , Its water absorption, water retention and fertility, and air permeability are good, and it is more suitable for the growth of seedling roots of flower seedlings. Therefore, mushroom dregs are more suitable for developing container seedling production substrates for flower seedlings. Therefore, the research and development g...
