Embodiment Construction

[0015] The present invention will be described in detail below in conjunction with specific embodiments.

[0016] Used equipment and reagent are as follows in the present embodiment: material: golden sunflower seed

[0017] Equipment: desiccator, 10mLEP tube, pipette, ear washing ball, Erlenmeyer flask, "Z" type tweezers, water bath, ultra-clean table, alcohol lamp, petri dish, constant temperature incubator.

[0018] Reagents: Concentrated hydrochloric acid, Clorox bleach, MS medium dry powder, 2,4-D (2,4-dichlorophenoxyacetic acid), 6-BA (6-benzylamino adenine or cytokinin), KT (hormone).

[0019] The specific culture method provided in the present embodiment is as follows:

[0020] 1. Seed disinfection: Soak the dried sunflower seeds in a 50°C constant temperature water bath for 30 minutes, then absorb the water on the surface of the seeds, and then disinfect them with chlorine gas for 4 hours.

[0021] The disinfection steps are as follows: first put the seeds to be ste...

Abstract

The invention provides a culturing method for sunflower callus tissue. The culturing method comprises the following steps that after a sunflower seed is soaked, water on the seed surface is absorbed up, and then the sunflower seed is disinfected with chlorine gas; the disinfected seed is inoculated on an aseptic seedling culture medium, and the aseptic seedling culture medium is placed in a constant-temperature dark environment to enable the seed to germinate after inoculation and then transferred to a constant-temperature greenhouse to continue to grow after the seed germinates; cotyledon and true leaves of an inoculated aseptic seedling are selected as explants and placed on an induction medium after the edges of the cotyledon and the true leaves are cut off, the surfaces of the cotyledon and the true leaves are gently pressed with blunt tweezers to enable the cotyledon and the true leaves to be closely attached to the surface of the induction medium, and an opening is sealed with two layers of paraffin wax sealing membranes; after being inoculated, the explants are placed in a constant-temperature biochemical incubator to be cultured until the callus tissue grows out; the callus tissue is cut off and transferred and inoculated into a subculture medium to continue to be cultured until the callus tissue grows to a stable period, and culturing is completed. According to the culturing method, the culturing cycle is shortened by utilizing the cotyledon of the aseptic seedling as the explant and the appropriate hormone concentration.

Description

technical field [0001] The invention provides a method for cultivating golden sunflower callus, belonging to the technical field of bioengineering. Background technique [0002] A large number of documents have proved that polyploid plants have higher yields on vegetative organs than diploid plants. Therefore, in order to obtain higher yields, scientific and technological workers often carry out polyploid breeding on plants and have achieved success, such as tetraploid strawberry and octoploid wheat. One of the difficulties in polyploid breeding is how to effectively establish a tissue culture system. The commonly used tissue culture of stem segments with buds has the disadvantages of heavy workload, low mutation rate, troublesome screening, and high mortality. Therefore, there is an urgent need in the art to provide a callus that can amplify small mutations through subculture and detect them through karyotype analysis, so as to perform polyploid breeding and cytological e...
