Problems solved by technology

[0004] At present, the application of polymer hydrogel in soft tissue repair has received extensive attention and research, but its composition and structure are complex, and there are also challenges such as mechanical strength, structure and comprehensive performance that cannot be unified and improved together, and need to be further improved: ①For physical Hydrogels that interact as cross-linking points are relatively weak in strength, which is not conducive to their clinical promotion and commercialization; ②For hydrogels constructed with c

[0039] The present invention also provides a method for preparing a double-crosslinked hybrid hydrogel based on acylhydrazone bonds and hydrophilic-hydrophobic self-assembly, which includes:

[0040] Step 110, preparing hydrazide-modified hyaluronic acid HAHZ;

[0041] Step 112, preparing aldehyde-modified polyethylene oxide-polyoxypropylene-polyoxyethylene PFAH;

[0042] Step 114, at room temperature, mix hydrazide-modified hyaluronic acid HAHZ with aldehyde-modified polyethylene oxide-polyoxypropylene-polyoxyethylene PFAH and water solvent in a preset ratio;

[0043] Step 116, stirring evenly to obtain a pre-crosslinked gel;

[0044] In step 118, the pre-crosslinked gel gradually becomes an elastic water-swellable three-dimensional network product or hydrogel.

[0045] Wherein, in step 114, HAHZ and part of the water solvent can be mixed to obtain a mixture 1, PFAH can be mixed with a part of the water solvent to obtain a mixture 2, and then the mixture 1 and 2 can be mixe...

Embodiment 1

[0052] Accurately prepare 5% (w / v) HAHZ and 5% (w / v) PFAH polymer solutions, mix the two thoroughly, and stand at room temperature to cross-link to form a gel.

Embodiment 2

[0054] Accurately prepare polymer solutions of 5% (w / v) HAHZ and 10% (w / v) PFAH respectively, add growth factors in the solution, mix thoroughly, and stand at room temperature to form a gel.

Abstract

The invention relates to the field of preparation of hydrogel and in particular relates to double-crosslinking hybrid hydrogel based on an acyl hydrazone bond and hydrophilic and hydrophobic self-assembling and a preparation method thereof as well as a skin tissue wound repairing agent. The double-crosslinking hybrid hydrogel is prepared from the following raw materials: hydrazide modified hyaluronic acid HAHZ and formyl modified polyethylene oxide-polypropylene oxide-polyethylene oxide PFAH; the adding mass ratio of the HAHZ and the PFAH meets the following conditions: the range of a hydrazide substitution degree/a formyl substitution degree is 20 to 80 percent and the balance is of a water solvent. According to the double-crosslinking hybrid hydrogel based on the acyl hydrazone bond andthe hydrophilic and hydrophobic self-assembling, advantages of covalent crosslinking and physical mutual effects are sufficiently combined and the prepared double-crosslinking hybrid hydrogel has an excellent self-healing property; the problem that medical gel dressing is easy to break due to an external force is easy to solve; the double-crosslinking hybrid hydrogel has certain temperature responsiveness and good mechanical performance and in-situ molding property ; the double-crosslinking hybrid hydrogel is very suitable for repairing nursing or treatment of skin tissue wounds.

Description

technical field [0001] The invention relates to the field of hydrogel preparation, in particular to a double-crosslinked hybrid hydrogel based on acylhydrazone bonds and hydrophilic-hydrophobic self-assembly, a preparation method thereof and a skin tissue wound repair agent. Background technique [0002] Polymer hydrogel is a three-dimensional network prepared by chemical or physical cross-linking of polymer components, which has certain mechanical strength and elasticity after water swelling. Polymers can be divided into two categories, natural and synthetic, according to their sources. Natural hydrophilic polymers include polysaccharides (cellulose, starch, alginic acid, hyaluronic acid, chitosan, etc.) and proteins (collagen, gelatin, polylysine, polyglutamic acid, etc.). Synthesized hydrophilic polymers include polyalcohols (polyethylene glycol), modified polyesters (polyurethane, hydrophilic modified polycaprolactone, polylactic acid, etc.), acrylic acid and its deriva...
