Problems solved by technology

Currently, autologous, allogeneic or xenogeneic collagenous connective tissue grafts and synthetic connective tissue grafts are commonly used clinically, but they have disadvantages such as nerve compression, insufficient closure, and unclear toxicity
Some biohydrogel products (fibrin glue, gelatin, collagen sponge, etc.) have also been used for dura mater sealing, but they suffer from insufficient tissue adhesion, difficulty in gel formation in wet environments, rapid degradation, excessive swelling and Troublesome preparation, etc.
However, the suture operation procedure is complicated and cumbersome, which increases the risk of surgery, and is gradually replaced by a safe and simple sealant.
Although many liquid or hydrogel tissue adhesives that can be applied in wet environments have been reported, most of them have disadvantages such as weak adhesion, poor mechanical matching with tissues, and excessive water absorption and swelling causing spinal cord compression.

Embodiment 1

[0032] A preparation method of a wet adhesive hydrogel for dural repair specifically comprises the following steps:

[0033] Step (1) Before preparing the reaction solution, use neutral alumina to filter acrylic acid, take 0.082 mL of acrylic acid in a single-neck round-bottom flask, add 50 mL of dichloromethane and mix, add 0.287 g of 1-(3-dimethylaminopropane) (base)-3-ethylcarbodiimide hydrochloride was fully stirred and activated at room temperature for 15 minutes, and then 0.173 g of N-hydroxysuccinimide was added and stirred at room temperature for 1 hour. Finally, 0.930 g of phosphatidylethanolamine was added and the mixture was fully stirred for 12 h at room temperature.

[0034] Step (2) The reaction solution of step (1) was vacuum distilled to concentrate the solvent, after adding an equal volume of anhydrous ethanol to the round-bottomed flask, a pale yellow precipitate was observed in the pale yellow solution, fully mixed and allowed to stand for 10 min. The organ...

Abstract

The invention relates to the technical field of medical materials, in particular to wet adhesive hydrogel for dura mater repair and a preparation method and application of the wet adhesive hydrogel for dura mater repair, and the preparation method comprises the following steps: S1, modifying an acrylic acid or acrylamide monomer with phospholipid to obtain the phospholipid modified acrylic acid or acrylamide monomer; and S2, mixing the phospholipid modified acrylic acid or acrylamide monomer prepared in the step S1 with acrylic acid, lauryl sodium sulfate, sodium lignin sulfonate, a cross-linking agent and a photoinitiator to prepare a hydrogel precursor solution, and polymerizing under ultraviolet irradiation to obtain the hydrogel. The preparation method comprises the following steps: grafting phospholipid with a hydrophobic chain to an acrylic acid or acrylamide monomer through an amidation reaction of carboxyl and amido, and preparing the hydrogel with swelling resistance and wet adhesion performance through a free radical polymerization reaction. The preparation method disclosed by the invention is simple to operate, and the prepared hydrogel has good wet bonding capacity and mechanical property, and can be effectively used for repairing dura mater and preventing leakage of cerebrospinal fluid.

Description

technical field [0001] The invention relates to the technical field of medical materials, in particular to a wet adhesive hydrogel for dura repair and a preparation method and application thereof. [0002] technical background [0003] Dura tear (DT) is one of the common complications after spinal surgery. Cerebrospinal fluid leakage caused by dural rupture can cause secondary symptoms such as headache, nausea, and vomiting, and even lead to serious consequences such as spinal cord infection and meningitis. The overall incidence of DT during spinal surgery is 3.8%-7.6%, and the incidence of postoperative CSF leak is 1%-17%. Under normal circumstances, small-scale dural defects are usually closed with sutures, and large-scale defects need to be repaired with dural patches combined with sutures or sealants. Currently, autologous, allogeneic or xenogeneic collagenous connective tissue transplantation and synthetic connective tissue transplantation are commonly used clinically,...
