Problems solved by technology

However, at this stage, traditional wound dressings cannot have all the above characteristics at the same time; the self-healing performance of the dressing needs to be realized with the help of dynamic chemical bonds, and dynamic chemical bonds are inherently unstable and do not have the characteristics of elastic recovery. This is the technical limitation of self-healing dressings at this stage

Embodiment 1

[0027] Example 1: Preparation of quaternized / N-methacryloyl chitosan and four-arm benzaldehyde-capped polyethylene glycol

[0028] (1) Preparation of quaternized / N-methacryloyl chitosan: the chitosan powder of 2g is completely dissolved in 100mL volume percentage concentration of 1% acetic acid aqueous solution, and the mass percentage concentration is 2 % chitosan acetic acid solution. 0.84g of methacrylic anhydride is slowly added dropwise to the above-mentioned chitosan acetic acid solution, and the reaction is continued for 12 hours under the conditions of heating and stirring in a water bath at 60°C, and the N-acylation reaction of chitosan can be completed. The pH value of the reaction mixed solution was adjusted to 7.0, and transferred to a dialysis bag, and dialyzed in a deionized water environment for 4 days, and the dialyzed solution was freeze-dried to obtain spongy N-methacryloyl chitosan. Dissolve 1.5 g of the above-mentioned spongy N-methacryloyl chitosan in 100...

Embodiment 2

[0030] Example 2: Preparation of hydrogel with self-healing, excellent elastic recovery and antibacterial properties

[0031] The prepared quaternized / N-methacryloyl chitosan and four-arm benzaldehyde-capped polyethylene glycol were respectively dissolved in water to obtain corresponding aqueous solutions, the concentrations of which were 15 mg / mL and 200 mg / mL respectively. Then the water-soluble blue light photoinitiator (lithium phenyl 2,4,6-trimethylbenzoyl phosphate) was dissolved in the quaternized / N-methacrylylated chitosan aqueous solution at a concentration of 30 μg / mL. The two solutions that have been prepared above are uniformly mixed according to the volume ratio of 1:4, and a self-healing hydrogel can be formed in a short time (within 1 minute). Then the formed self-healing hydrogel was irradiated with blue light (50mW / cm 2) for 30 seconds, triggering the cross-linking of carbon-carbon covalent bonds in the hydrogel, and finally a hydrogel with self-healing, ex...

Embodiment 3

[0032] Example 3: Performance evaluation of hydrogels with self-healing, excellent elastic recovery and antibacterial properties

[0033] The prepared columnar self-healing, excellent elastic recovery and antibacterial properties of two hydrogels loaded with different dyes were cut from the middle with a scalpel blade and divided into two. Then they were docked with hydrogels of different colors in pairs, and placed in a humid environment at 36°C for spontaneous healing for 120 minutes. Finally, use tweezers to pick up the healed hydrogel to check its border healing and whether it can fully support its own gravity without breaking. The study found that after simple physical docking and short-term (120 minutes) treatment, the re-self-healed hydrogel can fully support its own gravity without breaking, and the healing interface has obvious fusion phenomena, such as image 3 shown.

[0034] The self-healing, excellent elastic recovery and antibacterial properties of the prepared...

Abstract

The invention relates to a preparation method of a self-healing, excellent elastic recovery and antibacterial hydrogel dressing, the hydrogel mainly comprises quaternized / N-methylacryloylated chitosan and four-arm benzaldehyde end-capped polyethylene glycol, and blue light irradiation is combined for further crosslinking to prepare the multifunctional hydrogel. The prepared hydrogel has self-healing performance (dynamic covalent bonds), excellent elastic recovery (carbon-carbon double bond crosslinking) and antibacterial performance (quaternary ammonium salt modification), is used for repairing and regenerating skin tissue defects, and has outstanding application prospects in the direction of infected wound healing dressings. According to the present invention, the material has characteristics of rapid self-healing ability, excellent elastic recovery, good mechanical strength, and excellent antibacterial ability. The hydrogel is simple in preparation and operation process and low in production cost, and can be used as an effective novel wound infection dressing in treatment of chronic infection wounds at joints.

Description

technical field [0001] The invention belongs to the field of tissue engineering and regenerative medicine, and in particular relates to a self-healing, excellent elastic recovery and antibacterial hydrogel dressing and a preparation method, especially for repairing and regenerating skin tissue defects. Background technique [0002] Chronic wounds can develop from a variety of conditions—including diabetic neuropathy (diabetic foot ulcers), decubitus ulcers (pressure ulcers), and vascular disease (venous leg ulcers). Because chronic wounds are difficult to heal spontaneously and easy to relapse, chronic wounds are extremely susceptible to pathogenic bacteria infection, eventually resulting in non-traumatic amputation or even death, bringing great pain and challenges to patients and even the entire economy and society. Due to the large population base in my country and the increasing prevalence of diabetes, the treatment of chronic refractory wounds has become an important cli...
