Problems solved by technology

The most suitable raw material for itaconic acid fermentation is glucose, but the price of glucose relative to carbon sources such as starch makes the production cost increasingly unable to meet the actual industrial production
On the other hand, the itaconic acid-metabolizing bacterial strain lacks sugar-hydrolyzing enzymes or has low sugar-hydrolyzing enzyme activity, making the strains unable to effectively utilize macromolecular sugars (starch, etc.)

Abstract

The invention discloses an aspergillus terreus variant KY-AT-002 high in yield of itaconic acid. The aspergillus terreus variant KY-AT-002 is preserved in the China General Microbiological Culture Collection Center, the preservation address is No. 3, NO. 1 yard, Beichen West Road, Chaoyang district, Beijing, Institute of Microbiology, Chinese Academy of Sciences, the preservation date is June 3th,2020, the classification designation is aspergillus terreus, and the preservation number is CGMCC NO.19907. The aspergillus terreus strain KY-AT-002 high in yield of itaconic acid is identified as anaspergillus terreus variant through morphology analysis, ITS rDNA sequence analysis, complete genome re-sequencing analysis, and protein coding gene beta-Tubulin, Calmodulin and RPB2 sequence similarity comparison analysis. When the strain is subjected to shake flask fermentation culture, the inventor finds that the yield of the itaconic acid by the aspergillus terreus variant can reach 77.15g / L,and the sugar conversion rate reaches 60.89%; then the aspergillus terreus variant is subjected to amplified culture by a 50L fermenter, the yield of the itaconic acid can reach 84.21g / L, the sugar acid conversion rate reaches 66.82%, and the yield is stable. The strain has great industrial application prospects.

Description

technical field [0001] The invention belongs to the field of biological technology, and relates to a microbial strain and its application, in particular to the high-yiaconic acid-yielding Aspergillus terreus variety KY-AT-002 and its application. Background technique [0002] Itaconic acid is the fifth largest organic acid in the world (the first four are: citric acid, gluconic acid, lactic acid and malic acid). It is recognized by the US Department of Energy as one of the 12 high value-added chemical platform compounds derived from biomass. Biological fermentation is the main method of itaconic acid source. The itaconic acid strain currently used in industrial production is mainly Aspergillus terreus. Although smut, Candida and Rhodotorula also have the ability to secrete itaconic acid, the itaconic acid production is poor compared with Aspergillus terreus. So far, Aspergillus terreus is still the most widely used itaconic acid industrial production strain. The most suit...
