Embodiment 1

[0043]This example is used to illustrate how to prepare a fermentation broth using the Bacillus megaterium strain provided by the present invention.

[0044] 1. Raw materials

[0045] Incline medium: peptone 10g / L, yeast powder 5g / L, sodium chloride 10g / L, agar 15-20g / L, pH 7.2-7.4.

[0046] Fermentation medium: yeast extract 1-3g / L, peptone 0.5-2g / L, sucrose 0.5-2g / L, ammonium sulfate 0.5-1g / L, ferrous sulfate 0.1-1g / L, pH 6.7-7.2.

[0047] 2. Preparation method:

[0048] Pick the slant strains into the fermentation medium, fill the volume of 100mL / 500mL Erlenmeyer flask, and culture at 37°C and 180rpm for 20h with shaking.

[0049] 3. Detection of cellulase activity in fermentation broth

[0050] (1) Detection method of cellulase activity (refer to NY 609-2002 organic material decomposing agent)

[0051] a. Reagents and solutions

[0052] Substrate solution: Accurately weigh 0.625g of carboxymethylcellulose sodium salt, dissolve it in 100.0mL of sodium phosphate buffer ...

Embodiment 2

[0072] This embodiment is used to illustrate the application of the fermented liquid or culture liquid prepared in Example 1 in the decomposing of crop straws,

[0073] 1. Decomposition target:

[0074] Corn stalks are crushed to 3-5 cm, and the water content is about 35%.

[0075] 2. Method

[0076] 2.1 Preparation of microbial agent carrier

[0077] The food waste collected by the food waste collection and transportation vehicle is processed in the following steps:

[0078] (1) Use a rewinder to collect kitchen waste;

[0079] (2) Use vibrating grid sorting machine (patent number: ZL200910085142.1) for mechanical sorting: put the sorted kitchen waste liquid into comprehensive sewage treatment, put the sorted paper products, wood products, etc. into incineration , put the sorted food processing solid waste and food residue into the next step of processing;

[0080] (3) crushing and extruding the material obtained in step (2) by using a bag-breaking distributing machine u...

Embodiment 3

[0093] The difference between this embodiment and embodiment 2 is that the decomposing target is different. The decomposing target of this embodiment is garden waste: from landscaping trees and shrub branches and leaves, which are crushed to 3-5 cm and have a moisture content of about 35%.

[0094] Operate according to the same method, finish decomposing in 10 days, and the germination rate index of cabbage seeds is 91.45%. For the determination method of cabbage seed germination rate index, refer to DB11 / T 840-2011 technical regulations for landscaping waste composting.

Abstract

The invention belongs to the field of microorganisms, and in particular discloses a microbial strain BW-1701 for efficient degradation of cellulose and an application of the microbial strain. The microbial strain BW-1701 is bacillus megatherium which is preserved by preservation number of CGMCC No.15079. The strain is strong in capacity of generating cellulase, and the generated cellulase, which is kept at quite high activity, is applicable to fermentative degradation of raw materials rich in cellulose such as crop straws, garden wastes and the like. The strain, when applied to fermenting treatment of the crop straws and the garden wastes, can accelerate a decomposition speed and enhance a decomposition degree, and organic fertilizer can be generated within a short time and good fertilizerefficiency can be achieved. Straw burning, resource waste and environmental pollution can be relieved, and the utilization rate of wasted biological resources can be improved; and the microbial strain has obvious economic and ecological meaning.

Description

technical field [0001] The invention belongs to the field of microorganisms, and in particular relates to a microbial strain for efficiently degrading cellulose and its application. Background technique [0002] China is a big agricultural country, owning 7% of the arable land on the earth, and the output of crop straw is quite considerable, reaching 600 million tons per year. In the past, crop stalks could be used as fodder to feed livestock; as fuel for heating and cooking; as materials for building houses; as fermented manure and returned to the fields. However, with the increasing popularity of agricultural mechanization, less and less crop straw is used to feed livestock, heat cooking and build houses, and more will be burned on site. Burning straw will not only cause waste of resources, loss of organic matter, N, S, but also form thick smoke, affect aviation safety, and damage the atmospheric environment. Therefore, it is imperative to find a healthy, environmentally...
