The method comprises the steps of: carrying out induction of the protocorm and enrichment culture of the protocorm by using iron sheet dendrobe seed, stem tip or root tip as explant, wherein the enrichment culture medium components are as follows: every 1L of 1/2MS culture medium contains 100 mL of coconut milk, 0.2 mg of NAA, 1.0 mg of KT, 30 g of cane sugar, 0.3 mg of sodium selenite, 0.35 mg of chromium trichloride and 8 g of agar; pH is 5.6-5.8. The embryo-germinated protocorm, which is subjected to induction culture for 30-40 days and is as large as a sesame grain, is transferred to the enrichment culture medium, each bottle contains about ten grains of protocorm; and the protocorm is cultured under conditions that the temperature is 25 DEG C, the illumination intensity is 1500-2000 lux and the illumination time is 1-16 h/d.
