The application discloses a mutant of iridoid synthetase with improved specificity and enzyme activity and application thereof. The mutant of iridoid synthetase is obtained by mutation of wild-type iridoid synthetase NmISY2 derived from Nepeta mussinii. The mutant has higher catalytic activity to 8-oxogeranial, and the enzyme activity to geranial is obviously decreased. The ratio of the enzyme activity of the mutant to 8-oxogeranial and geranial is up to 293 times, which is greatly improved compared with the parent iridoid synthetase. The mutant can be used for heterologous synthesis and in-vitro synthesis of iridoid compounds and derivatives, and has a good application prospect.
