Application of sequential combination of CAR T based on improvement of tumor microenvironment in preparation of anti-tumor drugs

ActiveCN119971054BHydroxy compound active ingredientsAerosol deliveryProtein-Tyrosine KinasesCalcipotriol
The application discloses a sequential anti-tumor strategy based on improvement of tumor microenvironment (TME) combined with CAR T and application thereof, and belongs to the technical field of biological medicine, which simultaneously improves tumor fibrosis and acid microenvironment by applying tumor-related fibroblast (CAF) targeting drugs and proton pump inhibitors, and sequentially injects CAR T combined therapy for solid tumors, wherein the CAF targeting drugs are selected from tretinoin, calcipotriol, losartan and minnelide, the proton pump inhibitors are selected from lansoprazole, omeprazole, pantoprazole, rabeprazole and esomeprazole, and the CAR T receptor or ligand is selected from mesothelin, protein tyrosine kinase 7, NKG2D, CD47, B7-H3, MUC1 and HER2; the sequential administration strategy first destroys the dense physical barrier of the tumor microenvironment, improves the acidity of the tumor site, ensures the infiltration, survival and proliferation of CAR T at the tumor site, and then targets and inhibits the growth of solid tumors, especially high-malignancy triple-negative breast cancer, by using the specificity of CAR T.

A reaction kettle for processing rabeprazole sulfide

This utility model relates to the field of reaction vessel technology, specifically a reaction vessel for processing rabeprazole sulfide. It includes a reaction vessel body, with three mounting seats fixedly connected to the outer wall of the reaction vessel body. A rotating shaft is fixedly connected to each mounting seat, and a support rod is rotatably connected to the rotating shaft. An attachment lug is fixedly connected to each support rod, and a limit rod is slidably connected to the attachment lug. Two arc-shaped grooves are provided on each mounting seat, and a limit block is fixedly connected to the outer side of each mounting seat. The end of the limit rod engages with a hole in the limit block. First, the reaction vessel body is installed in a designated position, and the end of the limit rod is no longer engaged with the hole in the limit block. Then, the support rod is rotated around the rotating shaft to change its angle. When the support rod rotates to a suitable angle, the end of the limit rod engages with another hole in the limit block, effectively limiting the support rod and quickly adjusting its tilt angle, thus enhancing the overall stability of the reaction vessel body.

Polycyclic ketone monooxygenase mutants and their use in the synthesis of chiral lazaroids

The present application provides a polycyclic ketone monooxygenase mutant, the amino acid sequence of which is obtained by mutation of the amino acid sequence shown in SEQ ID NO. 2, and discloses a method for asymmetric biocatalytic synthesis of a lrazole drug based on the mutant. The enzyme catalyst developed based on the present application can be used to asymmetrically catalyze the synthesis of (R)-rabeprazole, lansoprazole and other lrazole compounds under mild conditions using a whole cell or enzyme protein catalytic method, without the need for chemical catalysts, and is environmentally friendly, being a green biocatalytic synthesis route.
