Ractopamine is a feed additive, banned in many countries, to promote leanness in animals raised for their meat. Pharmacologically, it is a phenol-based TAAR1 agonist and β adrenoreceptor agonist that stimulates β₁ and β₂ adrenergic receptors. It is most commonly administered to animals for meat production as ractopamine hydrochloride. It is the active ingredient in products known in the US as Paylean for swine, Optaflexx for cattle, and Tomax for turkeys. It was developed by Elanco Animal Health, a division of Eli Lilly and Company.

The invention provides a group of oligonucleotide aptamers Apt-1, Apt-2 and Apt-3 which are capable of simultaneously identifying clenbuterol hydrochloride and salbutamol, an oligonucleotide aptamer CLB-2 which is capable of identifying the clenbuterol hydrochloride with high specificity, an oligonucleotide aptamer SAL-5 which is capable of identifying the salbutamol with high specificity and two oligonucleotide aptamers RAC-5 and RAC-6 which are capable of identifying ractopamine with high specificity. Through an SELEX (Systematic Evolution of Ligands by Exponential Enrichment) technology based on Fe3O4 magnetic nanoparticle separation, a random oligonucleotide library is immobilized on avidin-enveloped magnetic nanoparticles by virtue of a complementary chain of a biotinylation marker, and the oligonucleotide aptamers, which are high in specific affinity, are finally obtained by conducting screening by 16 turns. The aptamers are broad in application prospect; and the aptamers, by virtue of marker function genes or fluorescent dyes, are applicable to detection of the clenbuterol hydrochloride, the salbutamol and the ractopamine in food; therefore, a new choice is provided for existing detection methods which depend on antibodies.

The invention relates to a novel synthesis method for a multilayer protection hyperstable water-soluble single fluorescent quantum dot and a fluorescent microsphere. The method comprises the following steps: transferring an oil-soluble fluorescent quantum dot to a water phase by using an amphoteric oligomer; then wrapping a water-soluble single fluorescent quantum dot and a fluorescent microsphere in silica; allowing the surfaces of the single fluorescent quantum dot and the fluorescent microsphere wrapped by silica to carry hydrophobic groups; and finally preparing the multilayer protection hyperstable water-soluble single fluorescent quantum dot and the fluorescent microsphere by using the amphoteric oligomer again. With the method, the fluorescent quantum dot can be protected from a destructive effect of an external environment (e.g., a strong acid, a strong alkali, a salt, etc.) to a greatest extent. The multilayer protection hyperstable water-soluble single fluorescent quantum dot and the fluorescent microsphere can be used in a microporous immunostrip biological detection system for detection of human chorionic gonadotropin (HCG), human immunodeficiency virus (HIV), hepatitis and clenbuterol--ractopamine(Rac) residual and the like in swine urine.

The invention is composed of a scale board, a sample pad, a magnetic combination pad, a coated film, an absorbent pad and the sample pad; the magnetic combination pad, the coated film and the absorbent pad are connected one by one on the scale board. The magnetic combination pad is made from absorbed glass mat which is used to absorb ractopamine magnetic antibodies; detection line T which is printed by the carrier protein solution coupled with ractopamine and control line C which is printed by goat anti-rabbit IgG are positioned on the coated film. The preparation method comprises the following steps: inserting the sample pad of the test paper in the sample solution to be tested, taking out the sample pad after 10-20 seconds, reacting at room temperature for 15 minutes, testing the test paper in a magnetic signal detector which outputs the biological response signals converted to magnetic field signals in the form of electrical signals; drawing standard curve and then evaluating the ractopamine content in the sample to be tested according to the standard curve. Preparation method and application of ractopamine molecularly-imprinted materials

The invention discloses a preparation method of ractopamine molecularly-imprinted materials, and is characterized by comprising the following steps: (1) the activation of silica gel: weighing silica gel, and carrying out backflow activation on the silica gel by using an acidic aqueous solution; (2) the double-bond bonding of the silica gel: after the activated silica gel and a double-bond containing silane coupling agent are heated, and carrying out a reaction on the two; (3) the surface chemical grafting of the silicon gel: putting the silica gel into a polyethylenimine aqueous solution to carry out a heating reaction to obtain an adsorbing material; and (4) product preparation: adding the adsorbing material into a ractopamine aqueous solution, adjusting the system pH value of the obtained mixture to an alkaline value, adding a crosslinking agent in the alkaline mixture, and carrying out a heating reaction on the alkaline mixture; and after the heating reaction is completed, washing off template molecule ractopamine, and drying the obtained object so as to obtain a ractopamine molecularly-imprinted material. Compared with the prior art, the preparation method disclosed by the invention has the advantages that the method is simple in preparation process, strong in maneuverability and low in preparation cost; and the prepared molecularly-imprinted material is good in specificity, short in adsorption equilibrium time to ractopamine, short in elution time and good in elution effect.
