Hydrangea macrophylla is a species of flowering plant in the family Hydrangeaceae, native to Japan. It is a deciduous shrub growing to 2 m (7 ft) tall by 2.5 m (8 ft) broad with large heads of pink or blue flowers in summer and autumn. Common names include bigleaf hydrangea, French hydrangea, lacecap hydrangea, mophead hydrangea, penny mac and hortensia. It is widely cultivated in many parts of the world in many climates. It is not to be confused with H. aspera 'Macrophylla'.

The method includes the steps of: 1) selecting healthy and robustly growing and disease-free hydrangeamacrophylla seedlings, scissoring newly growing branches, removing leaves, retaining parts of petioles, and performing cleaning for standby use; 2) conducting immersion disinfection on the branches with an alcohol solution and a mercuric chloride solution in order on a super clean workbench, and rinsing the branches clean; 3) cutting the branches into small pieces, inoculating the small pieces ofbranches into a medium, and conducting culture in a culture room; 4) after culturing for a period of time, sprouting axillary buds, cutting off the sprout tender branches, and inoculating the branchesinto a proliferation medium for culture; 5) after culture for a period of time, forming a small amount of calluses and adventitious buds at the root, dividing the plants into several clusters, inoculating the plants into a clustered seedling induction medium for culture, further inducing adventitious buds and rooting to form multi-branch rooting seedlings, and putting the seedlings in a greenhouse for acclimatization; 6) after acclimatization for a period of time, cleaning the seedlings, and conducting seedling hardening in a full-illumination spray greenhouse; and 7) at the end of seedling hardening, planting the seedlings in a 1 gallon container for next-step cultivation, thus forming a full-plant container seedling product. The rapid cultivation method for hydrangea macrophylla container seedlings provided by the invention eliminates the pinching process, has short cultivation period, does not affect the flowering of the next year, has high consistency, and can achieve full plants,and is suitable for large-scale and factory anniversary production of hydrangea macrophylla container seedlings.

Rapid propagation method of Hydrangea macrophylla

The invention belongs to the technical field of forestry vegetative propagation, and particularly relates to a rapid propagation method of Hydrangea macrophylla. The method comprises the following steps: (1) carrying out sterile disinfection treatment by taking a stem section of the Hydrangea macrophylla as an explant; (2) cutting the Hydrangea macrophylla stem section treated in the step (1) intoHydrangea macrophylla stem sections each with at least one axillary bud, inoculating the Hydrangea macrophylla stem sections into a culture medium, and germinating the axillary buds; (3) when the axillary buds in the step (2) extend to a certain length, cutting off the axillary buds, inoculating the axillary buds into a culture medium, and carrying out proliferation on the axillary buds to obtaina large number of axillary buds; (4) inoculating the axillary buds in the step (3) into a culture medium, and carrying out induction culture to form rooting seedlings; and (5) transplanting tissue culture seedlings in the step (4), namely the rooting seedlings, into a matrix, and carrying out culture to obtain Hydrangea macrophylla aseptic seedlings with robust rhizomes. The method has the advantages of being high in propagation speed and large in propagation coefficient, and is not limited by seasons and propagation materials.

The method provided by the invention has the characteristics of high survival rate, short propagation period, a multiplication coefficient up to 6.2, and a rooting rate up to 93%, completes rooting and seedling hardening in one step, shortens the seedling raising time by 1-2 months, can realize anniversary generation of dwarf hydrangea macrophylla, and has broad market prospects.
