Hypophthalmichthys is a genus of large cyprinid fish consisting of three species. The name comes from Greek ὑπό, (hypó) "below"; ὀφθαλμός (ophthalmós), "eye"; ἰχθῦς (ichthŷs), "fish", thus "fish with eyes below", referring to the fact that the fish has its eyes below the mouth line. Members of this genus are native to fresh water in East Asia, ranging from Siberia to Vietnam, but have been widely introduced outside their native range, where often considered invasive.

The application belongs to the technical field of fish breeding, and discloses a method for knocking out PHD3 gene of Hypophthalmichthys molitrix and application thereof in creating new germplasm of low-oxygen-tolerant Hypophthalmichthys molitrix. Taking PHD3 gene as a target gene, two gRNAs targeting the first exon of PHD3 gene are synthesized by in vitro transcription, mixed with Cas9 protein, and used to edit genes of wild-type Hypophthalmichthys molitrix, so as to improve the knocking-out efficiency of PHD3 gene, and then F0 generation mutants of Hypophthalmichthys molitrix with PHD3 gene knocked out are efficiently obtained. The F0 generation mutants of Hypophthalmichthys molitrix with PHD3 gene knocked out are subjected to low-oxygen stress experiment, and the result shows that the low-oxygen tolerance of the PHD3 knockout mutants of Hypophthalmichthys molitrix is significantly improved compared with the wild type. The application successfully realizes efficient knocking out of PHD3 gene of Hypophthalmichthys molitrix and obtains Hypophthalmichthys molitrix PHD3 mutants with significantly enhanced low-oxygen tolerance, thereby providing an effective technical means for carrying out functional research and genetic improvement of Hypophthalmichthys molitrix, and having important industrial application value.
