By using a split-PE design and a pegRNA architecture processed by hCtRNA, the TRU-PE system achieves simultaneous editing of up to 10 gene loci, with the editing efficiency being improved by up to 20 fold, effectively expanding the application range in hard-to-transfect cell types and complex gene loci. The gene editing system overcomes the application bottlenecks of prime editing technology in terms of size, delivery efficiency and multiplex editing. Moreover, the system is compatible with a variety of prime editor architectures and has wide applicability.
