A method for simultaneously preparing modified stevia dry leaves and crude steviol glycoside based on glucose

PendingCN122405767AFood supplementStevia <genus>
This invention provides a method for the simultaneous preparation of modified stevia leaves and crude glucosyl steviol glycosides, belonging to the field of food additives. The invention first involves disrupting the cell walls of stevia leaves using a plant cell wall-degrading enzyme, then directly adding a glycosyl donor using cyclodextrin glucosyltransferase as a catalyst in a one-pot glycosylation reaction. This allows for the simultaneous release and modification of steviol glycosides, achieving a final conversion rate of 68.57%. After the reaction, the mixture is filtered and freeze-dried, simultaneously obtaining modified stevia leaves with intact morphology and surface and pore interior loaded with modified glucosyl steviol glycosides, as well as crude glucosyl steviol glycoside powder. Sensory evaluation of the modified products showed that the modified stevia leaves exhibited significantly better sweetness, bitterness, and astringency than the unmodified leaves, and the crude glucosyl steviol glycosides had a significantly better taste than the extract from the unmodified stevia leaves. This invention features a short process flow, low cost, and achieves full utilization of stevia raw materials, demonstrating promising prospects for industrial application.
