Radiosurgical neuromodulation devices, systems, and methods for treatment of behavioral disorders by external application of ionizing radiation

Radiosurgical techniques and systems treat behavioral disorders (such as depression, Obsessive-Compulsive Disorder (“OCD”), addiction, hyperphagia, and the like) by directing radiation from outside the patient toward a target tissue within the patient's brain, typically without imposing surgical trauma. The target will often be included in a neural circuit associated with the behavioral disorder. A cellularly sub-lethal dose of the radiation may be applied and the radiation can mitigate the behavioral disorder, obesity, or the like, by modulating the level of neural activity within the target and in associated tissues. Hypersensitive and / or hyperactive neuronal tissue may be targeted, with the radiation downwardly modulating hyperactive neuronal activity. By down-regulating the activity of a target that normally exerts negative feedback or a limiting effect on a relevant neural circuit, the activity of the circuit may be increased.

Methods and substances for directed RNA editing

PendingUS20190093098A1Accumulation of mutationAltered levelHydrolasesPolymorphism usesDiseaseNucleotide
The invention relates to methods and substances for the targeted alteration of genetic information on an RNA level. The substances are artificially produced guide RNAs, which are capable of recruiting endogenous editing enzymes, such as hADAR enzymes, in particular hADAR2 and hADAR1, in order to introduce targeted point mutations in selected mRNAs. The guide RNA consists of multiple segments and is constructed in such a way that individual nucleotides from different segments pair to form a double helix, and the nucleotides of a determined segment form a hairpin structure within the guide RNA. The invention also relates to the method for directed RNA editing, wherein the guide RNA is transfected into the cells in which the RNA editing is to be carried out. The substances and the method can be used for repairing individual, e.g. disease-relevant point mutations, such as those leading to premature stop signals. An advantage of the invention is that endogenous editing enzymes are also used in order to introduce targeted point mutations into the RNA. Only the short guide RNA, used for recruiting endogenous editing enzymes, must be artificially produced for each specific problem and ectopically expressed.

System for active toner concentration target adjustments and method to maintain development performance

A closed-loop toner concentration adjustment system extends xerographic process control performance with a system and method including a first controller to adjust toner concentration (TC) in a developer of the imaging device to alter a DMA level of the imaging device, the TC being adjusted based on a TC target value; a second controller to adjust an electrostatic development field to alter the DMA level; and an adjustment logic device to adjust the TC target value based on adjustments made to the electrostatic development field. The first controller outputs a dispense amount to the developer based on a difference between a measurement of the actual TC and the TC target value, thereby adjusting the TC based on the dispense amount to the developer. The second controller adjusts the electrostatic development field based on a difference between a measurement of the actual DMA level and a DMA target value. The adjustment logic monitors the adjustments made to the electrostatic development field to determine a trend, and adjusts the TC target value based on the trend.

Binary Passive Variable Stiffness Joint

A variable stiffness joint and method to alter the stiffness of the joint with multiple stiffness levels is described wherein a plurality of stiffness bits (m) are used for enabling 2 m stiffness level variations for the joint. Each stiffness bit comprises an elastic element in mechanical connection with a clutch (21, 22, 23). The joint revolves with zero stiffness level when all the clutches (21, 22, 23) are disengaged whereas a clutch (21, 22, 23) involves one of the elastic elements which alter the stiffness of the joint. Engaging other clutches (21, 22, 23) involve more elastic elements for altering the joint stiffness and the resultant joint stiffness is determined by adding the stiffness values of all the involved springs (6, 7, 8).
