The method comprises the following steps: collecting soil light reflectance values on 10 signature wavelengths, wherein the characteristic wavelengths are 400nm, 730nm, 1026nm, 1296nm, 1544nm, 1885nm, 1912nm, 2013nm, 2205nm and 2314nm; converting the soil light reflectance values into soil absorbance values; calculating the content of the organic matters in the soil by the soil absorbance values of the characteristic wavelengths through a multiple linear regression equation. The device comprises a spectrum collection device and an embedded type processing device, wherein the spectrum collection device comprises a base, a sample box, a cylindrical rotary table and a bracket fixedly arranged on a base plate; the sample box is fixedly arranged on the upper surface of the base and the embedded type processing device is mounted inside the base; the rotary table is vertically arranged rightly above the sample box by the bracket. The lobular red sandalwood nursery stock cultivated by the cultivation method disclosed by the invention can grow to be used for making furniture about 20 years later after transplanting, and the growth rate of the lobular red sandalwood is significantly increased.
