The invention discloses a preparation method of mesenchymal stem cells capable of reversing
senescence. The preparation method comprises the following steps: S1, selecting and evaluating a donor; S2, separating human mesenchymal stem cells; S3, performing in-vitro proliferation culture on the cells; S4, performing
cell evaluation; S5, preparing a product; and S6, performing
cell cryopreservation. According to the invention, the activity of the cells in a mouse body is higher, the
cell death number is smaller, the preparation exclusion is smaller, and the characteristic promotes preparation cells to replace regional aging cells in the mouse body to realize reverse
senescence, and can also be used for injection treatment of diseases including diabetes, burns,
osteoarthritis, graft-versus-host diseases and the like. A serum-free culture medium is selected for in-vitro proliferation culture of the cells, and a
human platelet lysate is added into the serum-free culture medium to improve the
viscosity of the culture medium, so that the cells are prevented from mechanical damage, cell modification in the proliferation process is avoided, and potential risks existing in conventional fetal calf serum culture, for example,
comorbidity with
infection sources and microbial pollutants of people and
livestock, and immune response, are avoided.
