According to the method, (2S)-3-halogenated-1,2-propylene oxide and phosphoryl choline salt are used as the raw materials, and are subjected to esterification and hydrolysis reaction in a solvent to obtain the L-alpha-choline glycerophosphate crude product, and the crude product is subjected to silica gelcolumn chromatography and purification through ion exchange resin to obtain the L-alpha-choline glycerophosphate pure product. Compared with the prior art, the preparation method disclosed by the invention has the advantages that the operation is simpler; the adopted chiral material (2S)-3-halogenated-1,2-propylene oxide is cheap and easy to obtain; the prepared crude product is subjected to silica gelcolumn chromatography and purification through ion exchange resin to obtain the L-alpha-choline glycerophosphate product with higher quality, and the L-alpha-choline glycerophosphate content is more than 99%; the technical process is more suitable for large scale production.

Synthesis method of L-alpha-glyceryl phosphoryl choline

The embodiment of the invention discloses a synthesis method of L-alpha-glyceryl phosphoryl choline. The synthesis method comprises the following steps of: adding lecithin into a methanol solution, adding an alkali metal alkoxide catalyst into the solution, and performing hydrolysis reaction to remove an acyl group; regulating the pH of the solution to 5-6 by using an organic acid or an inorganic acid; extracting the solution after regulating the pH by using petroleum ether, and removing fat-soluble impurities in the solution; further removing inorganic salts and the impurities through strongly acidic, strongly alkaline, weakly acidic and weakly alkaline resin to obtain a sticky matter-like L-alpha-glyceryl phosphoryl choline crude product; and dissolving the sticky matter-like L-alpha-glyceryl phosphoryl choline crude product with methanol, adding acetone to precipitate crystals, and then performing suction filtration and drying to obtain a white solid finished product. The synthesis method disclosed by the invention has the advantages of simple process, low price of raw materials, easiness in acquisition of the raw material, simplicity in purification, low equipment requirements and easiness in implementation of industrial transformation.
