The invention provides a method for qualitatively or quantitatively detecting
urea in grains or distiller's
yeast, and belongs to the technical field of food detection. The
pretreatment method for detecting
urea in grain or distiller's
yeast provided by the invention comprises
urea extraction and derivation steps: the urea extraction step comprises the following steps: mixing a grain or distiller's
yeast sample with
activated carbon, a
zinc acetate solution and a
potassium ferrocyanide solution, carrying out ultrasonic treatment, filtering with medium-speed
filter paper, discarding 1-2mL of primary filtrate, and collecting subsequent filtrate as a sample solution; the
derivation method comprises the following steps: mixing a sample solution, a 9-hydroxyton solution and a
hydrochloric acid solution, placing in a dark place, and filtering to obtain a to-be-detected solution; according to the
pretreatment method provided by the invention, interference substances in the sample are removed through the steps of
activated carbon adsorption,
zinc acetate and
potassium ferrocyanide precipitation, the matrix effect is reduced, further, 9-hydroxyton is adopted as a
derivatization reagent and reacts with urea to generate a
fluorescent derivative, the extraction efficiency is high, and the detection sensitivity and accuracy are improved.
