The invention discloses a microRNA (Ribonucleic Acid) response type xanthene-hemicyanine fluorescent dye as well as a two-component probe system, a synthesis method and application thereof. The structure of the fluorescent dye is shown as a general formula I, and the fluorescent dye can be connected with a targeting ligand (such as short-chain RNA) through carboxyl. The two-component probe system is composed of two short-chain RNAs which are respectively connected with the dye and a biological orthogonal reaction partner (such as tetrazine), a biological orthogonal reaction is triggered through target microRNA-21 mediated hybridization, and the near-infrared fluorescence and photosensitive activity of the dye are specifically activated. The dye and the probe system can realize fluorescence imaging with high signal-to-noise ratio for overexpressed microRNA-21 in tumors, and generate active oxygen under laser irradiation for photodynamic therapy, thereby realizing integration of diagnosis and treatment.
